Is reduced SERCA2a expression detrimental or beneficial to postischemic cardiac function and injury? Evidence from heterozygous SERCA2a knockout mice

Abstract: Recent studies have demonstrated that increased expression of sarco(endo)plasmic reticulum Ca2+-ATPase (SERCA) 2a improves myocardial contractility and Ca2+ handling at baseline and in disease conditions, including myocardial ischemia-reperfusion (I/R). Conversely, it has also been reported that pharmacological inhibition of SERCA might improve postischemic function in stunned hearts or in isolated myocardium following I/R. The goal of this study was to test how decreases in SERCA pump level/activity affect ca… Show more

“…Cholesterol also can affect SERCA expression (58) and may accumulate excessively in CF lung (59). In one model, heterozygous SERCA2 knockout (SERCA2a 1/2 ) mice showed substantially enlarged infarction after cardiac ischemia (60). In addition, they showed impaired postischemic myocardial relaxation and reduced postischemic myocardial contractile function.…”

“…At day 14 after tumor/PBS inoculation, in vivo cardiac contractile function was evaluated by transthoracic two-dimensional (2D) and M-mode echocardiography (17). Echocardiography was performed using a GE Vivid7 ultrasound system (GE, Waukesha, WI) with a linear probe (Model i13L; frequency 14 MHz).…”

“…In this way, a SERCA2 deficiency would promote diastolic dysfunction and tachycardia. Studies of mice with a genetic deletion of one SERCA2 allele have demonstrated the importance of ER Ca 2ϩ uptake in protecting cardiac myocytes against ischemic injury in a model of myocardial infarction (Talukder et al, 2008).…”
