Is Reaction Acceleration of Microdroplet Chemistry Favorable to Controlling the Enantioselectivity?

Boyu Zheng,
Yikang Wu,
Luyun Xue
et al.

Abstract: Microdroplet chemistry has been proven to amazingly accelerate many chemical and biological reactions in the past 2 decades. Current microdroplet accelerated reactions are predominantly symmetric synthetic but minorly asymmetric synthetic reactions, where stereoselectivity is scarcely concerned. This study selected unimolecular and bimolecular reactions, multicomponent Passerini reactions, and enzymatic ketone reduction as the model reactions to illustrate whether reaction acceleration of microdroplet chemistr… Show more

“…In our previous studies, ,,,, both the reaction and collection temperature play essential roles in many microdroplet/thin film reactions, leading to a necessity to optimize the two temperatures. According to our investigation into the reaction temperature (entry 12 of Table and Table S4), high reaction temperatures were unbeneficial to the microdroplet/thin film conversion.…”

“…The extraordinary effect of nanoscale heating is continuously documented in all of these examples. Much like the unique reactivity observed when a reaction is run in a microdroplet, heating on the nanoscale is not well studied or understood enough to adequately hypothesize what unique effects it may produce. , Additionally, several thermodynamically feasible and operationally simple transformations have remained out of reach due to prohibitively high activation energy barriers. While theoretically possible, reactions have been abandoned because of prohibitive side reactivity or poor yields.…”

“…Parallel to us, the research group of Sun and Cheng was wondering about the same issues. 10 In their very recent publication, the authors analyzed the enantioselectivity in sonic sprayed microdroplets taking into account three different reaction cases. In the first one, the authors considered the retention of a chiral center in a C–N cleavage of a unimolecular reaction finding that pre-existing stereogenic centers of chiral starting materials are maintained in microdroplet conditions.…”
