IS 1236, a newly discovered member of the IS3 family, exhibits varied patterns of insertion into the Acinetobacter calcoaceticus chromosome

Abstract: Analysis of spontaneous mutations in Acinetobacter calcoaceticus revealed a 1237 bp insertion sequence named IS7236 and possessing a nucleotide sequence resembling those of members of the IS3 family. The chromosome of A. calcoaceticus strain ADPl contains seven copies of IS7236 which appears to insert preferentially into pobR, the transcriptional activator of the structural gene for p-hydroxybenzoate hydroxylase. IS7236 creates tandem 3 bp DNA duplications flanking the sites of its insertion in pobR. Different… Show more

“…IS1236 was first discovered in studies of mutations that prevent the degradation of p-hydroxybenzoate (18,19). The spontaneous insertion of IS1236 plays a significant role in the inactivation of genes for aromatic compound catabolism in two different chromosomal regions of ADP1 (18,19,26,41).…”

“…In A. baylyi ADP1, there is a single type of IS element, IS1236 (5,18,19,25). In the chromosome, there are six copies of IS1236, five of which are identical.…”

“…As example; the same expanded-spectrum beta-lactamase PER-1 gene was found in a chromosomally-integrated transposon in some A. baumannii isolates, and on a conjugative plasmid in others (Poirel et al, 2005); transposon Tn5613 of unknown function (but closely linked to various aromatic catabolic genes) is integrated into the chromosome of Acinetobacter sp. ADP1 (section 6.1.1) (Gerischer et al, 1996;Barbe et al, 2004); and conjugative transposon Tn2009 of A. junii 329 is located on the chromosome, is indistinguishable from a Streptococcus pneumoniae element (indicationg recent horizontal exchange), and can be transferred to various other Gram negative bacteria (Ojo et al, 2006).…”

“…The ADP1 genome has a small fraction of repetitive sequences (1.6%) ranging from a few short repeats to several complex ones. The longest repeated sequence is known as transposon Tn5613 (Gerischer et al, 1996;Barbe et al, 2004). Genome analysis has also revealed two principal prophage regions.…”
