Irisin and Secondary Osteoporosis in Humans

Abstract: Irisin is a peptide secreted by skeletal muscle following exercise that plays an important role in bone metabolism. Numerous experiments in vitro and in mouse models have shown that the administration of recombinant irisin promotes osteogenesis, protects osteocytes from dexamethasone-induced apoptosis, prevents disuse-induced loss of bone and muscle mass, and accelerates fracture healing. Although some aspects still need to be elucidated, such as the dose- and frequency-dependent effects of irisin in cell cult… Show more

“…The skeletal muscle secretes various molecules that affect bone including insulin-like growth factor-1 (IGF-1), basic fibroblast growth factor (FGF-2), interleukin-6 (IL-6), IL-15 and myostatin ( 39 ). Furthermore, irisin secreted by skeletal muscle was considered to improve bone health following exercise ( 40 ). Adipocytes also produce leptin, adiponectin and IL-6, which potentially modulate bone metabolisms.…”

“…The present research performed on young male volunteers during 10-day BR allowed us to analyze the circulating levels of molecules contributing to the physiological control of musculoskeletal system during and following adaptation to the change in mechanical load and fluid We studied the circulating levels of myokine irisin, whose anabolic role on bone and muscle has been recently demonstrated, [35][36][37] and of myostatin and sclerostin, well-known molecules controlling muscle and bone mass respectively. 38,39 Additionally, as an inflammatory state can arise as a result or outcome of lack of mobility, 40 we also measured the serum haptoglobin levels.…”

“…Linked with PGC-1α, exercise-induced myokines may also be used for treating MSD. Among them, there is large evidence linking irisin with bone health [85]. Indeed, irisin administration was reported to improve osteogenesis and fracture recovery, protecting from dexamethasone-induced cell death and disuse-induced bone loss [86].…”
