The A5 area at the ventrolateral pons contains noradrenergic neurons connected with other medullary areas involved in the cardiorespiratory control. Its contribution to the cardiorespiratory regulation was previously evidenced in anesthetized conditions. In the present study, we investigated the involvement of the A5 noradrenergic neurons to the basal and chemoreflex control of the sympathetic and respiratory activities in unanesthetized conditions. A5 noradrenergic neurons were lesioned using microinjections of anti-dopamine β-hydroxylase saporin (anti-DβH-SAP). After 7–8 days, we evaluated the arterial pressure levels, heart rate and minute ventilation in freely moving adult rats (280–350 g) as well as recorded from thoracic sympathetic (tSN) and phrenic nerves (PN) using the arterially perfused in situ preparation of juvenile rats (80–90 g). Baseline cardiovascular, sympathetic and respiratory parameters were similar between control (n = 7–8) and A5-lesioned rats (n = 5–6) in both experimental preparations. In adult rats, lesions of A5 noradrenergic neurons did not modify the reflex cardiorespiratory adjustments to hypoxia (7% O2) and hypercapnia (7% CO2). In the in situ preparations, the sympatho-excitation, but not the PN reflex response, elicited by either the stimulation of peripheral chemoreceptors (ΔtSN: 110 ± 12% vs 58 ± 8%, P < 0.01) or hypercapnia (ΔtSN: 9.5 ± 1.4% vs 3.9 ± 1.7%, P < 0.05) was attenuated in A5-lesioned rats compared to controls. Our data demonstrated that A5 noradrenergic neurons are part of the circuitry recruited for the processing of sympathetic response to hypoxia and hypercapnia in unanesthetized conditions.

The Locus coeruleus (LC) is a pontine area that contributes to the CO/pH chemosensitivity. LC cells express erythropoietin (Epo) receptors (EpoR), and Epo in the brainstem is a potent normoxic and hypoxic respiratory stimulant. However, a recent study showed that the intra-cisternal injection (ICI) of Epo antagonist does not alter the hypercapnic ventilatory response in mice. As ICI leads to a widespread dispersal of the product throughout the brainstem, in this work we evaluated the specific impact of Epo in the LC-mediated ventilatory response to CO (by whole body plethysmography) in juvenile male Wistar rats. Normocapnic and hypercapnic ventilation were evaluated before and after unilateral microinjection of Epo (1ng/100nL) into the LC. To evaluate the long-term effect of Epo, the HcVR was re-evaluated 24h later. Our results show that Epo attenuates the hypercapnic ventilation. We conclude that Epo in the LC tunes the hypercapnia-induced hyperpnea.

A Doença de Parkinson (DP) é caracterizada pela perda progressiva dos neurônios dopaminérgicos da Substância Negra porção compacta (SNpc). Estudos mostram redução de neurônios PHOX2B do núcleo retrotrapezóide (RTN) associada a um prejuízo da respiração em repouso e da resposta respiratória induzida por hipercapnia 40 dias após a indução da DP por injeção de 6-OHDA no estriado de ratos. No entanto, a deficiência respiratória induzida por hipercapnia foi restaurada 60 dias após a injeção de 6-OHDA. Nossa hipótese é que outros neurônios quimiossensíveis presentes no Sistema Nervoso Central (SNC) poderiam ser responsáveis pela restauração da quimiossensibilidade central nesse modelo de DP. Dessa forma, nosso objetivo foi investigar se os neurônios noradrenérgicos do Locus Coeruleus (LC), serotoninérgicos da porção pálida da rafe (RPa) ou orexinérgicos do hipotálamo lateral/região perifornicial (HL/PeF) seriam responsáveis por essa restauração nesse modelo. Em ratos, foi avaliada a ventilação (VE) em repouso, frente à hipercapnia (7% CO2) e hipóxia (8%O2) por até 60 dias após a indução do modelo de DP. Os animais foram subdividos em grupos que receberam veículo ou 6-OHDA no estriado e, além disso, também receberam veículo ou toxinas específicas para degenerar os neurônios do LC, da RPa ou do hipotálamo lateral/área perifornicial (HL/PeF). A 6-OHDA levou à redução de 86% dos neurônios da SNpc e as toxinas promoveram redução de 83% dos neurônios do LC, 90% dos neurônios da RPa e 78% dos neurônios do HL/PeF. Os resultados mostraram que nos animais com lesão da SNpc e do LC (n = 6) não houve recuperação da resposta ventilatória à hipercapnia, enquanto no grupo de animais com lesão da SNpc e da RPa (n = 8), a recuperação da resposta aos 60 dias permaneceu intacta; com relação aos animais com lesão da SNpc e HL/PeF (n = 6), observou-se uma recuperação da resposta à hipercapnia somente durante a fase escura e enquanto os animais estavam dormindo. Fizemos também experimentos para observarmos a ativação neuronal desses grupamentos durante hipercapnia e os dados mostraram redução do número de neurônios ativados por hipercania no RTN, e aumento no LC. Nossos dados sugerem que no modelo animal de DP pode ocorrer um processo de neuroplasticidade e os neurônios noradrenérgicos do LC são os principais candidatos a assumirem a função quimiossensível.
