Ionic Liquids as Matrixes for Matrix-Assisted Laser Desorption/Ionization Mass Spectrometry

Abstract: Room-temperature ionic liquids are useful as solvents for organic synthesis, electrochemical studies, and separations. We wished to examine whether their high solubalizing power, negligible vapor pressure, and broad liquid temperature range are advantageous if they are used as matrixes for UV-MALDI. Several different ionic matrixes were synthesized and tested, using peptides, proteins, and poly(ethylene glycol) (PEG-2000). All ionic liquids tested have excellent solubilizing properties and vacuum stability com… Show more

“…3 High molecular weight, 7 liquid, 8 inorganic and two-phase matrices 1 have all been utilised to overcome some of these problems. More recently, the use of ionic liquid matrices has become widespread 9 and the conditions required for the effective use of the matrix suppression effect (MSE) have been investigated. 10 The use of graphite as a matrix [11][12][13] and the utilisation of graphite target plates, without the addition of a standard crystalline matrix, 14 was discussed in our earlier publication, 15 where graphite powder and graphite rod were investigated as possible matrices for the analysis of actinides.…”

“…Similar to MALDI-MS applications, IL matrices were examined on the basis of ionization efficiencies as well as [159] and [163] signal enhancement. Using the best performing IL-based matrix, a high throughput ME-SIMS method that allowed for the analysis of 600 cells/h was developed with tandem MS for the detection and identification of metabolic signatures.…”

“…There have been many applications of ILs, including solvents for organic synthesis and liquidliquid extraction [1][2][3][4][5], gas chromatography stationary phases [6 -12] matrix assisted laser desorption/ionization (MALDI) matrices [13][14][15][16][17][18][19] and recently as ion pairing reagents for the trace analysis of anions by electrospray ionization mass spectrometry in the positive mode [20 -22].…”

“…These "hot spots" lead to poor shot to shot reproducibility. Other problems associated with solid matrices are difficult sample preparation, analyte size and/or type restrictions for different matrices [14], and harsh conditions used for preparation of the matrix [23]. Difficulty in sample preparation is typically caused by the analyte disrupting crystallization.…”
