In order to investigate the effect of potassium (K) nutrition on secondary metabolism, Catharanthus roseus was treated by different concentrations and forms of K nutrition (KNO 3 and K 2 SO 4 ). The results showed that the concentration of applied K obviously affected plant growth and biomass accumulation, while K form had little influence. The concentration of alkaloids, including vinblastine in the leaves and catharanthine in the leaves or roots, had a significant increase, especially upon 80 and 100 mM K nutrition. The measurement of internal ion content using energy dispersive spectroscopy displayed that the increase of K element in leaves was tightly correlated with elevated alkaloid concentration. The transcript levels of G10h, Str, and Dat genes, encoding key enzymes in the alkaloid biosynthesis pathway, were significantly activated by applying P and tightly related to alkaloid accumulation. In conclusion, our results provided the physiological and molecular evidence that potassium could effectively regulate alkaloid metabolism in C. roseus.

In a globalized economy, the control of fruit ripening is of strategic importance because excessive softening limits shelf life. Efforts have been made to reduce fruit softening in transgenic tomato through the suppression of genes encoding cell wall-degrading proteins. However, these have met with very limited success. N-glycans are reported to play an important role during fruit ripening, although the role of any particular enzyme is yet unknown. We have identified and targeted two ripening-specific N-glycoprotein modifying enzymes, α-mannosidase (α-Man) and β-D-N-acetylhexosaminidase (β-Hex). We show that their suppression enhances fruit shelf life, owing to the reduced rate of softening. Analysis of transgenic tomatoes revealed ≈2.5-and ≈2-fold firmer fruits in the α-Man and β-Hex RNAi lines, respectively, and ≈30 days of enhanced shelf life. Overexpression of α-Man or β-Hex resulted in excessive fruit softening. Expression of α-Man and β-Hex is induced by the ripening hormone ethylene and is modulated by a regulator of ripening, rin (ripening inhibitor). Furthermore, transcriptomic comparative studies demonstrate the downregulation of cell wall degradation-and ripening-related genes in RNAi fruits. It is evident from these results that N-glycan processing is involved in ripening-associated fruit softening. Genetic manipulation of N-glycan processing can be of strategic importance to enhance fruit shelf life, without any negative effect on phenotype, including yield.

Geraniol 10-hydroxylase (G10H) is a cytochrome P450 monooxygenase involved in the biosynthesis of iridoid monoterpenoids and several classes of monoterpenoid alkaloids found in a diverse range of plant species. Catharanthus roseus (Madagascar periwinkle) contains monoterpenoid indole alkaloids, several of which are pharmaceutically important. Vinblastine and vincristine, for example, find widespread use as anticancer drugs. G10H is thought to play a key regulatory role in terpenoid indole alkaloid biosynthesis. We purified G10H from C. roseus cells. Using degenerate PCR primers based on amino acid sequence information we cloned the corresponding cDNA. The encoded CYP76B6 protein has G10H activity when expressed in C. roseus and yeast cells. The stress hormone methyljasmonate strongly induced G10h gene expression coordinately with other terpenoid indole alkaloid biosynthesis genes in a C. roseus cell culture. ß
