Involvement of steroids in anti‐inflammatory effects of PK11195 in a murine model of pleurisy

Abstract: BACKGROUND: Studies on peripheral benzodiazepine receptor function have yielded a diverse list of activities of which the anti-inflammatory effects need to be further examined. AIMS: To evaluate the role of steroids, nitric oxide and adenosine-deaminase in the anti-inflammatory effect of PK11195. METHODS: Pleurisy was induced by intrapleural injection of carrageenan in mice pre-treated or not with PK11195. Leukocytes, exudation, adenosine-deaminase (ADA) activity and nitric oxide (NO) level were measured. Ster… Show more

“…Although Ro5-4864 presents an anti-inflammatory action very similar of that observed with PK11195 in a previous study (Da Silva et al, 2004), data obtained in both studies suggest that these two PBR ligands use distinct mechanisms. Namely, contrary to Ro5-4864, PK11195 inhibits neutrophil influx by a mechanism nonglucocorticoid dependent.…”

“…The TSPO is proposed to translocate cholesterol from the outer to the inner mitochondrial membrane for downstream synthesis of all steroids in the peripheral tissues, but in the CNS the end products are primarily neurosteroids (24)(25)(26). Earlier, TSPO ligands were shown to have antinociceptive and antiinflammatory effects (27,28 (29)(30)(31). Accordingly, we surmised that EETs activate TSPO and that the effects of synthetic sEHIs and natural EETs were mediated partially through an increase in the production of analgesic neurosteroids in the CNS.…”

“…Carrageenan-induced inflammation is a multicomplex phenomenon with the involvement of synthesis and/or liberation of a range of mediators such as prostaglandins, histamine, bradykinin, serotonin, nitric oxide, and leukotrienes and chemotaxis of neutrophils and macrophages [31]. After 24 h of carrageenan injection, there is an intense migration of leukocytes and an increase in levels of NO and cytokines [32][33][34][35][36].…”
