The present study assesses the resilience of the Mediterranean gilthead seabream (Sparus aurata) to acute warming and water acidification, using cellular indicators of systemic to molecular responses to various temperatures and CO2 concentrations. Tissue metabolic capacity derived from enzyme measurements, citrate synthase, 3-hydroxyacyl CoA dehydrogenase (HOAD), as well as lactate dehydrogenase. Cellular stress and signaling responses were identified from expression patterns of Hsp70 and Hsp90, the phosphorylation of p38 MAPK, JNKs and ERKs, from protein ubiquitylation and finally from the levels of transcription factor Hif-1α as an indicator of systemic hypoxemia. Exposure to elevated CO2 levels at temperatures higher than 24 °C generally caused an increase in fish mortality above the rate caused by warming alone, indicating effects of the two factors and a failure of acclimation and thus the limits of phenotypic plasticity to be reached. As a potential reason, tissue-dependent induction and stabilization of Hif-1α indicate hypoxemic conditions. Their exacerbation by enhanced CO2 levels is linked to the persistent expression of Hsp70 and Hsp90, oxidative stress and activation of MAPK and ubiquitin pathways. Antioxidant defence is enhanced by expression of catalase and glutathione reductase, however, leaving superoxide dismutase suppressed by elevated CO2 levels. On longer timescales in specimens surviving warming and CO2 exposures, various metabolic adjustments initiate a preference to oxidize lipid via HOAD for energy supply. These processes indicate significant acclimation up to a limit and a time-limited capacity to survive extreme conditions passively by exploiting mechanisms of cellular resilience.

This study aimed to identify and characterize the cellular and metabolic response in vital organs of Dicentrarchus labrax, after feeding on soy‐based diets enriched with different taurine concentrations (T0.2, T0.5, T1.0 w/w). Molecular responses (expression of Hsp70, Hsp90, phosphorylation of p38 mitogen‐activated protein kinase‐p38MAPK and extracellular signal‐regulated kinases‐ERK1/2) were addressed. Metabolic and antioxidant capacities such as activities of citrate synthase (CS), malate and lactate dehydrogenase (MDH, L‐LDH), superoxide dismutase (SOD), catalase (CAT) and levels of thiobarbituric acid‐reactive substances (TBARS) were assessed. The most intense changes were detected in the liver and in the anterior intestine showing that taurine induces important protective biochemical mechanisms. Specifically, in the liver and the anterior intestine, Hsp and MAPK levels, as well as L‐LDH activity levels were up‐regulated under the effect of taurine diets. T diet had no major effect on the activity levels of CS, SOD and TBARS, except for MDH in the liver and catalase in the anterior intestine, where activity levels of these enzymes were increased under the effect of T0.2 and T1.0, respectively. These results may facilitate monitoring of adequate health status of farmed fish, exploiting the beneficial T effects and could assist at the formulation of aquaculture feeding strategies.

Liver Transcriptome Changes in Zebrafish during Acclimation to Transport-Associated Stress

Liver plays a key role during the stress acclimation, and liver transcriptome analysis of shipped zebrafish could reveal the molecular adjustments that occur in the organ. Transcriptional changes in liver were analyzed with a 44 K oligo array using total RNA from fish prior to transport and during a mock transport process - immediately after packing (0 h), at 48 and 72 h. Large numbers of genes related to a variety of biological processes and pathways were regulated, mainly during transport (at 48/72 h). Immediately after packing, transcripts of genes related to both gluconeogenesis and glycolysis were induced. During transport, induction of gluconeogenesis-linked genes and reduction of glycolysis-related genes may be supporting the increase in blood glucose levels. Inhibition of genes involved in fatty acid beta-oxidation may be pointing to the poor ability of fish to utilize energy from fatty acids, under transport conditions. Genes involved in some of the mechanisms that regulate body ammonia were also affected. Even though genes associated with certain transaminases were inhibited in liver, sustained glutamate deamination may have led to high ammonia accumulation in liver/body. Enhanced levels of gene transcripts in ubiquitination and MAPK signalling cascade and reduced levels of gene transcripts related to ROS generation via peroxisomal enzymes as well as xenobiotic metabolism may be signifying the importance of such cellular and tissue responses to maintain homeostasis. Furthermore, transcripts connected with stress and thyroid hormones were also regulated. Moreover, suppression of genes related to specific immune components may be denoting the deleterious impact of transport on fish health. Thus, this study has revealed the complex molecular -adjustments that occur in zebrafish when they are transported.
