Involvement of lysosomal degradation in VEGF‐C‐induced down‐regulation of VEGFR‐3

Abstract: The vascular endothelial growth factor (VEGF)-C–induced down-regulation of VEGF receptor (VEGFR)-3 is important in lymphangiogenesis. Here, we demonstrate that VEGF-C, -D, and -C156S, but not VEGF-A, down-regulate VEGFR-3. VEGF-C stimulates VEGFR-3 tyrosyl phosphorylation and transient phosphorylation of ERK, p38, and c-Jun N-terminal kinases in lymphatic endothelial cells. VEGF-C–induced down-regulation of VEGFR-3 was blocked by a VEGF-C trap, tyrosine kinase inhibitor, and LPE (leupeptin, pepstatin, and E64)… Show more

“…Endostatin inhibits angiogenesis by directly binding to both VEGFR-1 and VEGFR-2 and blocking VEGF interaction with Flt-1 and Flk-1 to prevent VEGF-induced tyrosine phosphorylation of VEGFR-1 and VEGFR-2 and all downstream signaling events [102]. VEGFR-3 (also known as flt-4), another receptor tyrosine kinase, is expressed primarily on lymphatic endothelial cell surfaces [103]. Endostatin competitively inhibits VEGF binding to VEGFR-3 in vitro [104].…”

“…The corneal epithelium produces both pro- and anti-angiogenic factors. The secreted forms of VEGFR-1, -2 and -3 are also produced in the corneal epithelium [3033]. The VEGFR family is involved in both HA and LA, depending on the receptor subtype.…”

“…Endostatin binds both endogenous angiogenesis inhibitors thrombospondin-1 and SPARC (544, 545) and upregulates thrombospondin-1 expression (546). Endostatin also binds VEGFR2 on EC and VEGFR3 on lymphatic vessels preventing activation and downstream signaling (533,547,548). By occupying integrin-ECM binding sites, initiation of the tyrosine phosphorylation cascade, src activation, and EC migration are interrupted (549,550).…”
