Involvement of FGFR4 Gene Variants on the Clinicopathological Severity in Urothelial Cell Carcinoma

Abstract: Fibroblast growth factor receptor 4 (FGFR4) plays a prominent role in cell proliferation and cancer progression. This study explored the effect of FGFR4 single-nucleotide polymorphisms (SNPs) on the clinicopathological characteristics of urothelial cell carcinoma (UCC). This study was conducted to survey the possible correlation of the polymorphism of FGFR4 to the risk and clinicopathologic characteristics of UCC. Four loci of FGFR4 (rs2011077 T > C, rs351855 G > A, rs7708357 G>A, and rs1966265 A >… Show more

“…FGFR4 rs1966265 changes chemotherapy response in breast cancer [ 62 ], higher risk of oral squamous cell carcinoma susceptibility [ 31 ], initiation of cervical cancer (Taiwanese women) [ 19 ], and higher risk of breast cancer in Chinese women of Heilongjiang province [ 16 ]. FGFR4 rs2011077 TC+CC polymorphism is associated with higher tumor stage, tumor size, and grading in urothelial cell carcinoma [ 21 ]. FGFR4 rs2011077 with the GG genotype also increased the risk of prostate cancer in Japanese population [ 26 ].…”

“…The four SNPs investigated, namely rs2011077, rs351855, rs7708357, and rs1966265, in the FGFR4 genome region have been well defined and suggested to be associated with the risk of various cancers susceptibility [ 22 , 23 , 29 , 39 ]. The peripheral blood samples from lung cancer patients were collected for genomic DNA extraction.…”

“…Recent studies have focused on the clinical significance of FGFR4 genetic variations [ 10 ]. FGFR4 SNPs have been shown to be significantly associated with poor survival rates and worse prognosis in prostate cancer [ 11 , 12 ], head and neck cancer [ 13 , 14 ], breast cancer [ 15 , 16 , 17 , 18 , 19 ], ovarian cancer [ 20 ], hepatocellular carcinoma [ 21 ], uterine cervical cancer [ 22 ], urothelial cell carcinoma [ 23 ], pituitary tumors [ 24 , 25 ], soft tissue sarcoma [ 26 ], and oral squamous cell carcinoma [ 27 , 28 , 29 ]. However, the effects of FGFR4 SNPs on the risk of lung cancer seem to be controversial.…”

“…SNPs can exist in various regions of DNA and can affect the production of transcription factors, translation, and gene expression. Recently, the popularity and widespread use of SNP analysis platforms have made the identification of individual SNPs more convenient ( Tsay et al, 2019 ). The SNPs from FGFR4 have been recognized as an essential participant in cancer occurrence and were associated with prognosis in patients.…”

“…The SNPs from FGFR4 have been recognized as an essential participant in cancer occurrence and were associated with prognosis in patients. Examples are FGFR4 rs351855 in HCC ( Sheu et al, 2015 ), FGFR4 SNP rs2011077 with rs1966265 in urothelial cell carcinoma ( Tsay et al, 2019 ), and FGFR4 rs2011077 and rs1966265 in oral squamous cell carcinoma. FGFR4 can alter the production of relative transcript factors, which influences subsequent translation and gene expression ( Su et al, 2018 ) and is directly related to patient survival ( Chou et al, 2017 ).…”
