Investigation of the Symmetry of Oligomeric Enzymes with Bifunctional Reagents

Abstract: 1. The symmetry of proteins composed of identical polypeptide chains has been investigated by means of cross-linking with bifunctional reagents and subsequent sodium dodecylsulfate -polyacrylamide gel electrophoresis. The majority of the investigations were performed with diimidates of different chain lengths (C, -C,z), which react exclusively with amino groups. Aldolase, catalase, fumarase, pyruvate kinase, tetrameric proteins with identical polypeptide chains, reveal a D2 symmetry, i.e. they appear to be com… Show more

“…The analytical potential of these reagents in assessing the symmetry of tetrameric enzymes, for which X-ray diffraction data may not be available, has already been demonstrated [4]. Although the success of the method is clearly limited by the ease with which crosslinking can occur in any given system, the method has general applicability.…”

“…Only the three fractions of highest specific activity were pooled; there was a sharp fall in specific activity in the two fractions on the right-hand side of the peak. Sodium dodecyl sulphate/polyacrylamide-gel electrophoretograms of these fractions showed, in addition to the native arom complex band, eight to ten additional (Carpenter & Harrington, 1972;Hucho et al, 1975 molecular-weight standards in the gel-electrophoresis system used. From the resulting calibration (Fig.…”

“…It is of interest, taking into account recent data obtained from X-ray diffusion analysis [8], to examine if the crosslinking reaction with disucci~m~dyl ~-hydromuconate can be simulated by a simple model for the interactions between the polypeptide chains in an octamer built up of an equal number of 2 types of subunits. This model was calculated in a manner derived from that proposed [ 141 (6) and heptamers (7) to form an octamer (8) by theoretical intramolecular crosslinking of a protein containing 8 non-identi~ subunits. For the calculations, we assumed that the protein exhibits a pp,& structure in which only 3 types of different contact domains coexist (see insert).…”
