Investigation of the Regulation of Pigmentation in α‐Melanocyte‐Stimulating Hormone Responsive and Unresponsive Cultured B16 Melanoma Cells

Abstract: In vitro melanocyte-stimulating hormone (MSH) stimulates melanogenesis in some, but not all, melanocytes and melanoma cells. In an attempt to explain this variation in response to alpha MSH, we examined cyclic adenosine monophosphate (cAMP) accumulation, tyrosinase activity, and melanin production in primary (1 degree) murine B16 melanoma cells and in two B16 cell lines (B16 F1 and B16 F10) that are known to respond to alpha MSH. In vivo all three B16 melanoma cell types produced pigmented tumours. In vitro al… Show more

“…Thus, we evaluated melanogenesis activity on the basis of the sum of the amount of intracellular and extracellular melanin. According to the method of Hill et al, 13) the amounts of intracellular and extracellular melanin were determined separately. The sum of the amount of melanin was calculated by the addition of both average melanin amounts, and was represented without statistical standard errors.…”

“…The amount of melanin in cultured murine B16 melanoma cells (intracellular melanin) was measured according to the method of Hill et al, 18) as described in our previous paper. 19) Briefly, test samples were dissolved in DMSO/Ca Assay of Cell Proliferation The cell proliferation of murine B16 melanoma cells was assessed using the MTT method described in our previous paper.…”

“…A375 cells were seeded and treated as reported above, in order to measure extracellular and intracellular melanin according to the method of Hill et al (23). After 72 h of incubation, the culture medium was removed, centrifuged (700 x g, 10 min) and the supernatant was collected for extracellular melanin quantification.…”
