Investigation of the Hemostatic Effect of a Transdermal Patch Containing 0.55 mg Ethinyl Estradiol and 2.1 mg Gestodene Compared with a Monophasic Oral Contraceptive Containing 0.03 mg Ethinyl Estradiol and 0.15 mg Levonorgestrel: An Open-Label, Randomized, Crossover Study

Abstract: BackgroundTransdermal delivery of contraceptives offers several advantages over combined oral contraceptives (COCs), including effective absorption and the provision of relatively constant serum concentrations. Ethinyl estradiol (EE) and the progestin gestodene are well-absorbed through the skin and, therefore, well-suited for use in a transdermal contraceptive patch.ObjectiveThe objective of this study was to investigate the impact of a once-weekly transparent, transdermal patch delivering low doses of EE and… Show more

“…In 85.7% of participants, ovulation returned in the first cycle following termination of treatment 48. No alterations were observed in prothrombin fragments 1 + 2 or D-dimer when the patch was compared with a COC containing 0.03 mg EE/0.15 mg LNG 49. An application has been made to the European authorities; however, there is no information on whether an application has been made in the US.…”

“…In a recent study by Junge et al, 28 surrogate parameters for risk of venous thromboembolism were examined for a patch containing 0.55 mg of ethinylestradiol and 2.1 mg of gestodene. This was compared with an oral contraceptive pill containing 0.03 mg of ethinylestradiol and 0.15 mg of levonorgestrel.…”

“…Compliance in both groups was excellent. 28 Another pharmacological system, ie, a double-layer matrix releasing gestodene and ethinylestradiol steadily over one week, was introduced in 2009. 29 Overall, transdermal contraception can be offered as a safe, efficient, and convenient method of contraception.…”

“…[19][20][21] Finally, gonadal hormones are known to affect coagulation. [22][23][24] However, little of this has been evaluated after ICH. The first steps are (1) characterize gonadal hormone physiology after ICH and (2) define associations between hormone concentrations, pathophysiology, and recovery.…”
