Trehalose (Tre), which was an osmoprotective or stabilizing molecule, played a protective role against different abiotic stresses in plants and showed remarkable perspectives in salt stress. In this study, the potential role of Tre in improving the resistance to salt stress in tomato plants was investigated. Tomato plants (Micro Tom) were treated with Hoagland nutrient solution (CK), 10 mM Tre (T), 150 mM sodium chloride (NaCl, S), and 10 mM Tre+150 mM NaCl (S+T) for 5 days. Our results showed that foliar application of Tre alleviated the inhibition of tomato plant growth under salt stress. In addition, salt stress decreased the values of net photosynthetic rate (Pn, 85.99%), stomata conductance (gs, 57.3%), and transpiration rate (Tr, 47.97%), but increased that of intercellular carbon dioxide concentration (Ci, 26.25%). However, exogenous application of Tre significantly increased photosynthetic efficiency, increased the activity of Calvin cycle enzymes [ribulose diphosphate carboxylase/oxygenase (Rubisco), fructose-1,6-bisphosphate aldolase (FBA), fructose-1, 6-bisphosphatase (FBPase), glyceraldehyde-3-phosphate dehydrogenase (GAPDH), and transketolase (TK)], up-regulated the expression of genes encoding enzymes, induced stomatal opening, and alleviated salt-induced damage to the chloroplast membrane and structure. In the saline environment, photosynthetic electron transport was restricted, resulting the J-I-P phase to decrease. At the same time, the absorption, capture, and transport energies per excited cross-section and per active reaction center decreased, and the dissipation energy increased. Conversely, Tre reversed these values and enhanced the photosystem response to salt stress by protecting the photosynthetic electron transport system. In addition, foliage application with Tre significantly increased the potassium to sodium transport selectivity ratio (SK–Na) by 16.08%, and increased the levels of other ions to varying degrees. Principal component analysis (PCA) analysis showed that exogenous Tre could change the distribution of elements in different organs and affect the expressions of SlSOS1, SlNHX, SlHKT1.1, SlVHA, and SlHA-A at the transcriptional level under salt stress, thereby maintaining ion homeostasis. This study demonstrated that Tre was involved in the process of mitigating salt stress toxicity in tomato plants and provided specific insights into the effectiveness of Tre in mediating salt tolerance.

Vuralia turcica (Fabaceae; Papilionoideae) is a critically endangered endemic plant species in Turkey. This plant grows naturally in saline environments, although the photosynthesis and physiological functions of many plants are affected by salt stress.Molecular control mechanisms and identifi cation of genes involved in these mechanisms constitute the critical fi eld of study in plant science. Trehalose-6-phosphate synthase (TPS) is one of the essential enzyme genes involved in trehalose biosynthesis, which is protective against salt stress. Also, the vacuolar Na + /H + antiporter gene (NHX) is known to be useful in salt tolerance. In this study, the TPS and NHX-like genes in V. turcica were partially sequenced using degenerate primers for the fi rst time and submitted to the NCBI database (accession numbers MK120983 and MH757417, respectively). Also, the expression levels of the genes encoding TPS and NHX were investigated. The results indicate that the increase in both the level of applied salt and cadmium is coupled with the increase in the expression level of NHX and TPS genes. However, salt exposure signifi cantly affected the expression level of the NHX gene. The fi ndings suggest that the NHX gene might play a crucial role in the salt tolerance ability of V. turcica.

The Potential Effectiveness of Biochar Application to Reduce Soil Cd Bioavailability and Encourage Oak Seedling Growth

Today, it is very important to protect plants in soils contaminated with metals. We investigated the behavior of cadmium during the establishment of oak seedlings (Quercus castaneifolia C.A. Mey.) under biochar influence. This study was conducted in pots with loamy soil. Cadmium was added to soil at 0, 10, 30, and 50 mg per kg of soil, indicated by Control, Cd10, Cd30 and Cd50. Biochar was produced at 500–550 °C from rice husk and added at 1, 3, and 5% (wt/wt) levels, indicated by B1, B3, B5, and mixed with soil at planting in three replications. Generally, increasing biochar rates had significant effects on seedling height, diameter, and biomass. This coincided with Cd immobilization in the contaminated soil which reflects a decrease in Cd concentrations in the plant bioavailability of Cd. The tolerance index increased significantly, by 40.9%, 56%, and 60.6% in B1, B3, and B5 with Cd50, respectively, compared to polluted soil. The percent of Cd removal efficiency for Cd50 was 21%, 47%, and 67% in B1, B2, and B5, respectively. Our study highlights that biochar can reduce Cd bioavailability and improve the growth of oak seedlings in contaminated soil.
