Investigation of the Effect of Channel Structure and Flow Rate on On-Chip Bacterial Lysis

Abstract: Successful lysis of cells/microorganisms is a key step in the sample preparation in fields like molecular biology, bioengineering, and biomedical engineering. This study therefore aims to investigate the lysis of bacteria on-chip and its dependence on both microfluidic channel structure and flow rate. Effects of temperature on lysis on-chip were also investigated. To perform these investigations, three different microfluidic chips were designed and produced (straight, zigzag and circular configurations), while… Show more

“…AutoCAD 2022 (Autodesk, USA) software was then used to design the integrated hybrid chip and the shadow mask was produced from aluminium by Photofab (Cambridge, UK). The design of the chip and the fabrication procedure [40] are shown in figures 1(A) and (B). The integrated hybrid chip was fabricated on 25 mm by 25 mm glass slides by thermal evaporation (NANOVAK, Turkey) through a shadow mask at Bilkent University (UNAM, Turkey) (figure 1(B-I)).…”

“…For this, 5.85 g (1.00 mmol) of PDMS (component A) and 0.65 g (0.087 mmol) of crosslinker (Component B) was used for preparation of upper and bottom PDMS microchannels. The mixture of PDMS and crosslinker (9:1) was prepared by stirring for up to 3 min and added to molds and vacuumed in the desiccator for 20 min [32,33]. It was then left to stand at 50 °C overnight, and then the microfluidic chip was prepared by peeling off the PDMS layer.…”

“…3 Every year, about 3 million people with active TB are not diagnosed or notified under the national TB control program. 4,5 For this reason, drug susceptibility testing (DST) is significant to guide right treatment, but less than 1 in 20 new TB patients have access to DSTs. 6 Therefore, there is an urgent need for inexpensive, practical, and efficient diagnostic tools that can be used at the point of care.…”

“…The WHO reports that the overall detection of tuberculosis cases is still less than 60% in low‐income countries and only 66% worldwide 3 . For this reason, drug susceptibility testing (DST) is significant to guide right treatment, but less than 1 in 20 new TB patients have access to DSTs 6 .…”
