In the present study the potential toxic effect of prometryne herbicide was examined by investigating mitotic index, mitotic phase, chromosomal abnormalities, micronucleus frequency, 2C DNA content, and comet assay on the root tip cells of Allium cepa. The roots were exposed to 2, 3, 4 and 6 g l −1 concentrations within 12, 24 and 36 h. Mitotic index was markedly reduced by prometryne in each treatment group compared with the controls. The frequencies of mitotic phases have been changed. Prometryne significantly increased the anomaly cell percentage at all concentrations and application periods in comparison to their control. Mitotic abnormalities were recorded as disturbed prophase, c-mitosis, stickiness, laggards and chromatid bridges. The micronucleus was observed at interphase and its frequency was calculated in the test solutions used. Prometryne decreased the 2C DNA amount in onion root tips. Furthermore, the comet assay was applied to analyze the single stand breakages. Nearly all of the applied concentrations of prometryne enhanced DNA damage. In many cases negative correlation was found between 2C DNA amount and DNA damage; however, positive correlation was found between micronucleus formation and DNA damage.

SummaryThe present study was performed to evaluate the role of Aloe vera L. leaf extract (AvLE) on some physiological and cytogenetical parameters of Allium cepa L. seeds exposed to salinity. The radicle length of the seeds germinated in the medium with AvLE alone increased as compared with ones of the control seeds germinated in distilled water medium, while their radicle number and fresh weight reduced according to the control. In addition, the germination percentage of the mentioned seeds statistically was the same as the control seeds. Furthermore, the seeds germinated in the media containing 0.1 mg/L AvLE alone showed a significant increase of mitotic index, although they exhibited a higher number of chromosomal aberrations and micronucleus (MN) formation as compared with the seeds germinated in control conditions. On the other hand, salt stress considerably inhibited the seed germination and seedling growth of A. cepa. Moreover, salinity markedly decreased the mitotic index in root tip meristems of the seeds and increased the number of chromosomal aberrations. The negative effect of salt on the seed germination, seedling growth, mitotic activity and MN formation was alleviated in dramatically varying degrees by AvLE application. In contrast, the detrimental effects of salt on the chromosomal aberrations greatly increased with this treatment. Consequently, we reported that the application of AvLE alone induced chromosomal aberrations and MN formation, which are an indicator of genotoxicity, and had cytotoxic activity in normal conditions. However, the frequency of MN is greatly reduced in root tip cells of the seeds germinated in the medium containing 0.1 mg/L AvLE +0.15 M NaCl.

Detection of Cytotoxicity and Carcinogenicity of Soft Drink "Cola" on Allium Cepa Root Cells

The present investigation had been conducted to study the cytogenetic effects of soft drink "cola " at concentration (100%) with three exposure times (4, 6, and 8 hours) on root meristems of Allium cepa. The parameters tested were: mitotic index and chromosomal aberrations. Mitotic index was reduced in all exposure times 4,6 and 8 h with cola. On the other hand, total chromosomal abnormalities were highly increased in all exposure times comparatively with control. The physiological chromosomal aberrations observed were: (c-mitosis, stickiness, disturbance and laggard chromosome; and clastogenic aberrations were: chromosomal break and bridges. These findings indicate the cytotoxicity and carcinogenicity of the soft drink "cola". Our results supports the calls for warned for excessive intake of soft drinks especially "cola".
