Investigation of the aryl hydrocarbon receptor and the intrinsic tumoral component of the kynurenine pathway of tryptophan metabolism in primary brain tumors

Abstract: Collectively, these results suggest that AhR may offer a novel and robust therapeutic target for a patient population with highly limited treatment options.

“…Although the aberrant activation of KP and expression patterns of the first rate-limiting KP enzyme in gliomas have been previously reported, 5,9,10,13,[31][32][33][34] the systemic and comprehensive studies of the expression, activity, and individual function of IDO1 and its isozymes in gliomas are very limited.…”

“…Similar results have been observed for IDO1, IDO2, and TDO mRNA expression in the TCGA database 35 and IDO1/TDO protein expression of paraffin-embedded tissue sections from glioma patients. 13,31,32,34 Compared with IDO1/TDO, the function of IDO2 has been less studied. IDO2 plays a nonredundant role in Trp metabolism 36 and a nonredundant immunosuppressive role different from that of IDO1 and TDO.…”

“…The growth pattern of astrocytomas in the brain consists of invasive or local invasive growth, and as the invasiveness increases, the tumour grade increases and the survival rate decreases (10). At present, the conventional treatment is primarily surgical resection supplemented by radiotherapy and chemotherapy (11). There are a number of surgical methods for astrocytoma, including excisional biopsy (EB), subtotal resection (STR), gross resection (GR), partial resection (PR) and gross total resection (GTR).…”

“…IDO1 and TDO2 activity can lead to an immunosuppressed tumor microenvironment that promotes progression of glioblastoma (24). Production of KYN increases the occupancy of the aryl hydrocarbon receptor, thus enhancing the invasiveness of human glioblastoma cells (24,25).…”
