Investigation of the applicability of a sequential digestion protocol using trypsin and leucine aminopeptidase M for protein identification by matrix-assisted laser desorption/ionization - time of flight mass spectrometry

Abstract: An investigation of the applicability of a sequential digestion procedure involving endo- and exoprotease digestion of proteins is reported. The procedure involves the digestion of a protein sample with trypsin, yielding peptide fragments characteristic of the protein. The resulting mixture of peptide fragments is then subjected to N-terminal sequencing with leucine aminopeptidase M (LAP), with matrix-assisted laser desorption ionization-time of flight mass spectrometric analysis of the various digestion produ… Show more

“…In the pharmaceutical industry, inhibitors developed against these enzymes find application as drugs 7 . Aminopeptidases are also used for peptide sequencing 8 and in processing of recombinant proteins 9 . Today the industrial demand for aminopeptidases far exceeds their availability and the need to identify new sources is ever increasing 10,11 .…”

“…Analytical 2-D electrophoresis and in-gel proteolytic digestion were followed by Doucette and Li [14] and Gharahdaghi's method [15], respectively. The final tryptic digested peptides were resolved with 7 μl deionized water (DW) in 0.1% formic acid.…”

“…Here, we report on a new strategy designed to detect, without ambiguity, proteogenic peptide-like sequences in extra-terrestrial samples. For this purpose, we transposed a proteomics approach currently employed for the sequencing of peptides in biological samples using aminopeptidase M (APM, EC 3.4.11.2 16,17 ). In contrast to HCl hydrolysis which induces the release of amino acids from a wide variety of precursors, this exopeptidase catalyses stereoselectivity the stepwise removal of L--amino acids from the N-terminal end of peptides 18 (Figure 1).…”
