Abstract: Our data confirm that there is an inflammatory reaction in normal pregnancy compared with the non-pregnant state, which may be disrupted during miscarriage.

“…While moderate inflammation may be beneficial to pregnancy, excessive production of pro-inflammatory cytokines is harmful and contributes to adverse pregnancy outcomes, such as miscarriage, preterm birth (PTB) and preeclampsia (PE) (Redman and Sargent, 2004, Wei et al, 2010, Calleja-Agius et al, 2012. During placentation fetal trophoblasts form the growing placenta and invade the maternal uterine wall, interacting with maternal cells and modifying the uterine vasculature.…”

“…TNF-α has been shown to be increased in euploid compared to aneuploid miscarriages, as well as increased compared to the euploid pregnancies that survive [13,[72][73][74]79]. This increase has also been noted in their peripheral blood.…”

“…These cells also release IFN-γ to promote inflammation. It has been shown to be elevated in RPL patients [13,72,78,79,124]. Significant associations between RPL and the IFN-γ(+874) polymorphisms have been found [115,119,120].…”

“…Nevertheless there are conflicting reports regarding the inflammation state in early pregnancy loss, suggesting adequate balance for Th1/Th2 cytokines, although with a slight shift toward Th2 immunity in successful pregnancy (Saini et al, 2011). A recent study (Calleja-Agius et al, 2012) confirmed an inflammatory state (higher pro-inflammatory cytokines) in normal pregnancy compared with the non pregnant state, which may be disrupted during miscarriage. The study revealed in euploid miscarriage a shift toward Th1 immune response (higher TNF-/IL-6 ratio) at 6-9 weeks, but a lower TNF-/IL 10 and IFN-/IL10 ratios in the late first trimester compared to normal pregnancy.…”
