Investigation of sphingosine kinase 1 in interferon responses during dengue virus infection

Abstract: Dengue virus (DENV) regulates sphingosine kinase (SK)-1 activity and chemical inhibition of SK1 reduces DENV infection. In primary murine embryonic fibroblasts (pMEFs) lacking SK1 however, DENV infection is enhanced and this is associated with induction of normal levels of interferon beta (IFN-β) but reduced levels of IFN-stimulated genes (ISGs). We have further investigated this link between SK1 and type I IFN responses. DENV infection downregulates cell-surface IFN-alpha receptor (IFNAR)1 in both wild-type (… Show more

“…SK1 converts sphingosine to S1P, and when this is inhibited during DENV infection, although immune responses are largely unchanged, there are adaptive changes in IFNAR1 and IRF1 that compromise DENV-induced type I IFN responses (Aloia et al . 2017 ).…”

“…Viruses manipulate cellular signalling. Examples of viral exploitation of SphK/S1P are influenza [ 217 ], measles [ 213 ], hepatitis B [ 218 ], dengue virus [ 219 ], the respiratory syncytial virus (RSV), hepatitis B virus (HBV), and hepatitis C viruses (HCV) [ 220 ].…”

“…In addition to protein kinases, some lipid kinases have been targeted by antivirals. One example is sphingosine kinase 1, which affects replication of dengue virus (Aloia et al, 2017). Its activity can be determined by measuring the production of 32 P-labeled sphingosine-1phosphate from sphingosine and [γ 32 P]ATP (Clarke et al, 2016;Pitman et al, 2012).…”
