Investigation of protective effects of selenium and vitamin e against DNA oxidation, membrane damage and alteration of comt metabolism in smoke-exposed male mice ( Mus Musculus, Balb/c)

Abstract: In our study, the protective effects of vitamin E and Se (selenium) against cigarette smoke hazards on second-hand smoker (passive smoker) male mice (Balb/c) were investigated. Serum MDA levels in the smoke-exposed mice were found higher than serum MDA levels of control mice and Se- and vitamin E-treated mice. But, the MDA levels of smoke-exposed plus Se- and vitamin E-treated mice were found lower than MDA levels of smoke-exposed mice at the end of the three and five months. According to these results, applic… Show more

“…These effects might result from an alteration of the transport of isorhamnetin from tissue(s) into the circulation, a modification of the affinity of COMT for site-specific O-methylation of flavonol hydroxyl groups and/or an alteration in tamarixetin excretion from the liver into bile. An interaction between vitamin E status and COMT activity was recently suggested by a study in which vitamin E plus selenium prevented an increase in COMT activity induced by cigarette smoke in mice [54]. However, the mechanisms by which vitamin E status alters quercetin metabolism remain to be elucidated.…”

“…PM suppresses the intracellular levels of SAM, the methyl donor for DNMT via direct oxidation and inactivation of SAM by ROS [89]. In addition, PM up-regulates catechol-O-methyltransferase (COMT) [90], another methyltransferase utilizing SAM, which also degrades catecholamines such as dopamine, epinephrine, and norepinephrine [91]. …”

“…Formulation of effective and affordable antiulcer agents is an important goal in GI pharmacology, as the prescribed anti-ulcer drugs show various side effects and are expensive [3,4]. The trace element, selenium and some organo-selenium compounds can replenish the antioxidative defense, and are being used for treating gastric ulcers and cancers [5][6][7]. Very recently, a water-soluble selenium compound, DL-trans-3,4-dihydroxy-1-selenolane (DHS red ), synthesized by Iwaoka's group [8], could effectively heal the indomethacin (IND)-induced stomach ulceration in mice [9].…”
