Investigation of Fusion between Nanosized Lipid Vesicles and a Lipid Monolayer Toward Formation of Giant Lipid Vesicles with Various Kinds of Biomolecules

Abstract: We determined the properties of fusion between large unilamellar vesicles (LUVs) and the lipid monolayer by measuring the fluorescence intensity of rhodamine-conjugated phospholipids in cell-sized lipid vesicles. The charge of LUVs (containing cationic lipids) and lipid droplets (containing anionic lipids) promoted lipid membrane fusion. We also investigated the formation of cell-sized lipid vesicles with asymmetric lipid distribution using this fusion method. Moreover, cell-sized asymmetric ganglioside vesicl… Show more

“…The preparation of conventional floating lipid bilayers 15 is typically based on the Langmuir-Blodgett (LB) and/or Langmuir Schaefer (LS) techniques 9,41 . This approach obviously cannot be taken for the deposition of lipid bilayers onto fluid interfaces and the vesicle fusion technique 42,43 has to be employed instead.…”

“…In contrast, liposomes formed by the pulsed-jet flow method have long-term stability (up to one week). The flip-flop phenomenon and membrane protein reconstitution have been investigated on the asymmetric lipid vesicles formed by this method [ 64 , 65 ]. Modification of the double-well device allows for the formation of nano-sized asymmetric lipid vesicles [ 66 ], automatic formation of liposomes [ 67 , 68 ], and the formation of vesicles-in-a-vesicle structures that mimic the asymmetric lipid composition of plasma membranes and intracellular vesicle membranes [ 69 ].…”

“…The inkjet printing method starts from a planar, asymmetric bilayer formed at the contact of two aqueous droplets in an oily phase. From this bilayer, vesicles are ejected by the printing pulse [ 36 , 37 , 38 , 39 , 40 ].…”
