Investigation of Dynamic Behaviors of Asialoglycoprotein Receptors of Hepatocytes Using a Latex Probe Whose Surface Was Coated with Lactose-Carrying Polystyrene

Abstract: Characteristic properties of oligosaccharide-substituted poly styrenes as a hepatocyte-specific culture substratum were investigated from the viewpoint of adsorption and recognition. X-ray photoelectron spectra sug gest that the adsorption of the polymer onto a polystyrene film occurred in near mono-molecular layer thickness of 10-25 Å in globular conformations. Distri bution and migration of asialoglycoprotein receptors on the surface of a hepato cyte was followed under a scanning electron microscope using a … Show more

“…The representative XPS spectra of the polymer-coated substrates on the ODS-SAM are shown in Figure 7. 29 The C 1s spectra were resolved by the pseudo- Micropatterned Lectin Displays on the Glycoconjugate Polymer Substrates. The lectin recognition of the micropatterned carbohydrates was visualized by fluorescence microscopy using FITC-labeled lectin (Figure 8).…”

“…The synthesis of amphiphilic dextran grafted with polycaprolactone suitable for the preparation of biodegradable particles was also recently reported [54]. Various lowmolecular-weight saccharide (cellobiose [55], maltose [56], glucose [57], lactose [58])-containing monomers have been alternatively modified and used as reactive surfactants in emulsion polymerization of styrene. The resulting modified PS particles were also found to decrease protein adsorption as well as to serve as colloid supports for the covalent grafting of DNA probes using the reactivity of OH groups in anomeric position [59].…”

“…The biotinylated PAAm- 2) by the reaction with succinic anhydride: (A) before and (B) after the reaction. graft-PLLS (9) and not unlabeled copolymer (6) showed significant absorbance over the background level. The PAAm-graft-PLLS bound to the cells was, therefore, detectable by ELISA using the peroxidase-conjugated streptavidin.…”

“…We reported previously that the multivalent β-galactose ligands of a polystyrene derivative, poly[ N -( p -vinylbenzyl)-4- O -β- d -galactopyranosyl- d -gluconamide] [PVLA, poly(vinylbenzyl-β- d -lactonamide)], are recognized by the ASGP-R on the surface of the hepatocytes; namely, the PVLA is found to be an artificial cellular matrix as the useful substratum for hepatocyte culture ( ). When a culture dish is coated with a high density of PVLA, the hepatocytes attached on the dish remain spherical and show low DNA synthesis activity.…”
