Investigation of Combined Cyclodextrin and Hydrogel Formulation for Ocular Delivery of Dexamethasone Acetate by Means of Experimental Designs

Abstract: Dexamethasone acetate (DXMa) has proven its efficiency to treat corneal inflammation, without a great propensity to increase intraocular pressure. Unfortunately, its poor aqueous solubility, associated with a rapid precorneal elimination, results in a low drug bioavailability and a low penetration after topical ocular administration. The main objective of this study was to improve the apparent aqueous solubility of DXMa using cyclodextrins. First, hydroxypropyl-β-CD (HPβCD) and hydroxypropyl-γ-CD (HPγCD) were … Show more

“…The HPLC method was adopted as reported by Mazet et al and further validated. [17] The quantitative determination was carried out by HPLC (Agilent 1290, Agilent Technologies, USA) using reverse phase DikMa C18 column (50 × 4.6 mm, 5 µm) and a mobile phase composed of methanol and water (70:30, v/v) at a flow rate of 0.8 mL min −1 . The column temperature was at 25 °C, and chromatograms were obtained using a refractive index detector at a wavelength of 240 nm.…”

“…The drug quantification methodology was adapted from that previously reported [ 7 , 12 ] and validated in DXMa concentrations according to ICH Q2 (R1) guidelines in order to evaluate specificity, linearity, repeatability, intermediate fidelity, and limit of detection (LOD) and limit of quantification (LOQ) [ 13 ]. Quantitative determination were performed on a reversed-phase, high-performance liquid chromatographic (HPLC) component system LC 2010 AHT (SHIMADZU, Kyoto, Japan) consisting of a pump with degasser, an autosampler, a UV–VIS detector, and a column XTERRA ® MS C8, 5 µm particles, and 150 × 4.6 mm with a C8 cartridge.…”

“…Ethylene diamine–β-cyclodextrin encapsulation could hugely promote corneal curcumin delivery. On the other hand, Mazet et al [ 59 ] explored the effect of mixing dexamethasone with cyclodextrin to improve the aqueous solubility of curcumin. Hydroxypropyl-β-cyclodextrins and hydroxypropyl-γ-cyclodextrins were combined with a marketed mucoadhesive gel for topical administration.…”
