Investigation of Changes in Spermatozoon Characteristics, Chromatin Structure, and Antioxidant/Oxidant Parameters after Freeze-Thawing of Hesperidin (Vitamin P) Doses Added to Ram Semen

Abstract: We conducted this study to determine the potential cryopreservative effects of different hesperidin (vitamin P; H) doses on ram semen after freeze-thawing. Semen samples were obtained from Sönmez rams using an artificial vagina. The samples were divided into six groups: control, 10, 50, 100, 250, and 500 µg/mL H (C, H10, H50, H100, H250, and H500, respectively). At the end of the study, sperm motility and kinetic parameters, acrosome integrity (AI), mitochondrial membrane potential (MMP), viability, lipid pero… Show more

“…In addition, the freezing process negatively affects spermatological parameters and fertility (Kulaksız and Daşkın 2009). Due to all these factors, researchers focused on adding substances with high antioxidant properties to extenders during semen freezing (Yeni et al 2022). Capsaicin (8-methyl-N-vanilyl-6-nonenamide) is an active ingredient responsible for the pungency of hot peppers.…”

“…Supporting the DNA integrity result identified in the JB25 and JB50 groups in this study, it was demonstrated that 2, 4 and 6 μg/ml spirulina extract had a protective effect on spermatozoon DNA integrity, and a toxic effect when > 6 μg/ml of it was used (Mizera et al 2019). The findings of this study were in favour of the findings of the studies conducted by our researcher group in recent years, where we have used various antioxidants (rosmarinic acid, pine bark tree extract, vaccenic acid, hesperidin, thymoquinone) due to their remedial effects on DNA integrity (Yeni et al 2018;Taşdemir et al 2020;Avdatek et al 2022;İnanç et al 2022;Yeni et al 2022). It was concluded that as an antioxidant, JB at appropriate doses (25 and 50 μg/ml) maintains the integrity of DNA and this antioxidant property of JB manifests its healing effect on DNA damage by creating an additive effect along with other cryoprotectants in the extender.…”

“…However, an excess concentration of ROS during the semen freezing process causes oxidative stress (Gao et al, 2007). The resulting oxidative stress brings with it undesirable effects that may even include the loss of motility in the sperm cell (Yeni et al, 2022). In this study, it was determined that all doses of SA, except for the 0.5 mM dose, had an improving effect on progressive and total motility.…”
