Breast cancer continues to be the most frequently occurring cancer in women around the world. For women, the three most commonly diagnosed cancers are breast, lung and bronchus, and colorectal. Representing one-half of all the cases; breast cancer alone is expected to account for 30% all new cancer diagnoses in women. An estimated 41,070 breast cancer deaths will occur in 2017 (Siegel et al., 2017). In India, it is the cancer of breast alone which is expected to cross the figure of 200000 by the year 2021 (Takiar et al., 2010). Based on the stage of diagnosis, breast cancer is treated with a multidisciplinary approach involving surgery, radiation and systemic therapy including chemotherapy or hormonal therapy (Harfindal and Helms, 2006). Surgery alone may increase the chances of relapse so combinations of chemotherapeutic agents are given. The side effects of chemotherapy depend on the individual, the drug used, the schedule and dose used. These side effects can include fatigue, risk of infection, nausea, vomiting, mouth sores, hair loss, anorexia, diarrhea and bone marrow suppression. Hormonal modifiers like Tamoxifen are given in estrogen

Pleurotus eryngii (P. eryngii), also known as the king oyster mushroom, contains many bioactive substances, including polysaccharides, lipids, peptides, sterols and triterpenoids. The aim of this study was to explore the antitumor and immunomodulatory activities of P. eryngii extract. The fresh P. eryngii material was extracted with MeOH (methanol) and then partitioned with EtOAc (ethyl acetate). The study showed that the P. eryngii EtOAc extract (EAe) not only has antitumor effects by in vitro and in vivo methods but also significantly increases some immune activities. In addition, histopathological observations showed that EAe‐treated mice exhibited much lower cancerization throughout the liver and lung. EAe can also promote antioxidant activity, which may regulate the body's antioxidant capacity and activate the antitumor mechanism. Practical Applications Our study revealed the anticancer and immunomodulatory activity of Pleurotus eryngii extract, and identified the key components of the P. eryngii extract with HPLC‐MS. These findings explain the antitumor activity of P. eryngii, which is very useful for developing the potential antitumor drugs for clinical application.

This study aims to investigate the effect of Phyllanthus niruri Linn (Euphorbiaceae) in the proliferation of human leukemic cells (MOLT-4 and K562). Phyllanthus niruri L (P.niruri) was macerated by using various solvents to obtain the crude extracts. Citotoxicity of the extracts against MOLT-4 and K562 cells was tested using MTT assay to find the IC50 value. To analyse cell cycle progression, cellular DNA was measured using propidium iodide (PI) staining. Apoptosis induction was evaluated using Annexin V-FITC and PI staining and was analysed using FACSVerse flow cytometry. Finally, the expression of p53 on MOLT-4 and K562 cell lysate was measured using western blotting, to identify the possible mode of action of anticancer activity. P. niruri crude extracts demonstrated a potential anticancer effect towards MOLT-4 cells (IC50 range was 42.21±4.98 to 97.06±18.29µg/mL). However, against K562 cells, P.niruri extracts exhibited a lower inhibitory potency (the IC50 was 120.19±8.48 to 256.55±26.22µg/mL). The results showed the selectivity of the toxic effect of the extracts against MOLT-4 and K562. To evaluate the possible mechanism of action of the anticancer effect, we evaluated P. niruri extract action in apoptosis induction and p53 expression. The results showed that methanol and hexane extract inhibited MOLT-4 cell progression from G1 to S-phase, indicating G1 cell arrest. Moreover, apoptotic cell population following treatment of MOLT-4 and K562 cells with methanol extract was markedly increased, showing morphological signs of apoptosis including membrane degradation and chromatin condensation. Furthermore, we found that there was an increase in p53 expression following MOLT-4 treatment with methanol extract, suggesting that p53 induction might be involved in cell apoptosis. The results indicated the involvement of p53 pathway in the mechanism of anti-cancer activity exerted by P. niruri extract on MOLT-4 cells. However, on cancer cells lacking P53 expression, such as K562 cells, apoptosis might take place via other pathways.
