SummaryThe occurence of Armillaria species was assessed in Norway, enabling the northern-most distribution of this genus to be determined in Europe. Four Armillaria species were found in Norway. Armillaria borealis was the most common species occurring on woody vegetation to the permafrost zone (ca. 69°N). Armillaria cepistipes was present in southern and central Norway, but was not found further than 66°N. Armillaria solidipes and Armillaria gallica were rare, found at only one locality each; 59°40¢ and 59°32¢, respectively. Armillaria species were found on 14 hosts, but there was no significant difference between occurrence of A. borealis and A. cepistipes on declining and dead trees. Phylogenetic analyses separated each species into separate clades. All isolates of A. borealis, except one, and most isolates of A. solidipes were in separate clades. However, a subclade within the A. borealis clade was formed of two A. ostoyae and one A. borealis isolates. Two small A. cepistipes genets were found in a declining oak stand.

DNA sequences (~3 kb long) extending from the intergenic spacer 1 (IGS1) region to the 18S gene were obtained for isolates of Armillaria ostoyae, Armillaria calvescens, Armillaria gallica, and Armillaria sinapina. Additional investigation of 16 A. ostoyae, 11 Armillaria gemina, 21 A. calvescens, 18 A. gallica, and 15 A. sinapina isolates produced 117 sequences spanning the 3' end of the IGS1 through the 5S gene and into the 5' end of the IGS2 region. Additional sequences spanning the 3' IGS2 to 5' 18S gene region were obtained for two A. ostoyae, three A. gemina, two A. calvescens, two A. gallica, and three A. sinapina isolates. This is the first report of complete IGS2 sequences from Armillaria spp. A species identification protocol involving species-specific primers and restriction fragment length polymorphism analysis was devised based on species-specific polymorphisms. The protocol successfully identified all 16 A. ostoyae, 11 A. gemina, three of three Armillaria mellea, 18 A. gallica, 14 of 15 A. sinapina (11/12 diploid and 3/3 haploid), and 14 of 21 A. calvescens (13/15 diploid and 1/6 haploid) isolates included in this study. To the best of our knowledge, this success rate has not been matched by other methods.

Populations of Armillaria species in pine plantations in west-central Poland

Two diploid isolates of Armillaria gallica and 143 diploid isolates of A. solidipes were obtained from 145 samples of rhizomorphs, fruit bodies and infected wood of sessile oak and Scots pines from 5-10-year-old Scots pine plantations in three Forest Districts located 100-350 km apart in west-central Poland. Based on pairings among the 145 isolates, 18 somatic compatibility groups (genets) of A. solidipes were distinguished in the three plantations. Sequencing of ITS1/2 rDNA of 18 isolates representing the 18 genets delineated four nuclear haplotypes. This suggests that many of the A. solidipes genets are closely related and were possibly established by sib-related basidiospores. With a few exceptions there was general geographical specialization of genets but not haplotypes. Geographical specialization of genets and the dominance of single haplotypes suggest that the A. solidipes population results from clonal rather than sexual reproduction. Sequencing of the ITS1/2 and IGS-1 rDNA showed small nucleotide diversity in ITS1/2 rDNA and much more diversity in IGS-1 rDNA of the isolates of A. solidipes studied. However, none of these regions has sufficient resolution for the clear differentiation of A. solidipes from A. borealis. The sequences of the EF 1-alpha gene showed high interspecific variability in Armillaria species and very low intraspecific variability in A. solidipes. This gene is the most appropriate for reliable identification of biological species and subgroups of Armillaria. Its application is the most useful in ecological and epidemiological studies of Armillaria.
