Investigating the influence of perinatal nicotine and alcohol exposure on the genetic profiles of dopaminergic neurons in the VTA using miRNA–mRNA analysis

Abstract: Nicotine and alcohol are two of the most commonly used and abused recreational drugs, are often used simultaneously, and have been linked to significant health hazards. Furthermore, patients diagnosed with dependence on one drug are highly likely to be dependent on the other. Several studies have shown the effects of each drug independently on gene expression within many brain regions, including the ventral tegmental area (VTA). Dopaminergic (DA) neurons of the dopamine reward pathway originate from the VTA, w… Show more

“…We identified several similarities across cigarette smoking and alcohol use traits. For example, neuronal processes such as neuronal migration and apoptosis were associated with cigarette smoking and alcohol use, which is in line with studies that have pinpointed the disruption of neuronal migration and neurotransmission in response to substance use 2931 . We also observed myelination and gliogenesis to be associated with DPW and CPD, respectively, hinting at the role of neuron-glia interactions in substance use traits 32, 33 .…”

“…As shown in Figure 1b, the running time of calculating the feature importance using the Random Forest model increases linearly against the feature size. The short running time facilities the generation of stable feature The classification accuracy of the algorithm (i.e., correctly predicting all three labels) was 68 ± 15 %, which was similar to other classification accuracy of other samples from our studies [29,31]. This method is rather stable.…”

“…All experiments were performed in accordance with the protocols approved by the Institutional Animal Care and Use Committee (IACUC) and the University of Houston Animal Care Operations (ACO). The detailed protocol has been published previously [29,31,32]. Briefly, pregnant Sprague-Dawley (SD) rats (Charles River, Wilmington, MA, USA) were maintained at standard conditions and were given an ad libitum diet.…”
