Invasive Properties of Histone Transformed Cells

Abstract: Histone preparationChromatin was isolated from the livers of adult rats (Scott-Russ strain) according to the method of Bonner et al. (1968), omitting centrifugation through dense sucrose solution. Histone was extracted from the isolated chromatin by blending with 0-25N HCI. The insoluble material was centrifuged off and the extraction procedure repeated. The supernatants were pooled and histone was precipitated from the acid extract by adding 9 volumes of acetone. The precipitate was washed with acetone, air d… Show more

“…However, recent observations made in this laboratory suggest that this opinion is not entirely valid. For example, we have been able to show that instead of a loss of viability in BHK21 cultures challenged with crude rat liver histone, there is an increase in cell activity and invasiveness in in vitro conditions (Latner, Longstaff and Lunn, 1971). In the present communication, we provide evidence to show that BHK21 cells, similarly treated with rat liver histone, are also invasive in vivo.…”

“…The invasion of mouse (Bar Harbor strain 129) kidney cortex explants by polvoma virus transformed hamster kidney cells (BHK21/C13/Py, Macpherson and Stoker, 1962) was investigated using the mixed cell/ organ culture technique developed by Latner et al (1971). Monolayers of cells were grown in the filter wells of the apparatus and kidney explants cultured above them.…”

“…The bh oad spectrum protease inhibitor aprotinin (Trasylol, Bayer) was selected for study because it is known to accunmulate rapidly in the renal tissues when admiinistered intravenously (Haberland, 1967 (Latner et al, 1971).…”

“…Various attempts to investigate the process in vitro have achieved some success (Easty and Easty, 1963;Barski and Wolff, 1965;Yarnell and Ambrose, 1969a, b;Latner, Longstaff and Lunn, 1971) but have not been susceptible to quantitative analysis. The methods used have frequently involved the confrontation of tumour cells with the cut edges of normal tissues.…”
