Invasive cervical cancers from women living in the United States or Botswana: differences in human papillomavirus type distribution

Abstract: BackgroundCervical cancer is the primary cause of cancer-related deaths in women living in Botswana.MethodsParaffin-embedded blocks of formalin-fixed invasive cervical cancer specimens were identified from women living in the U.S. (n = 50) or Botswana (n = 171) from which DNA was extracted. Thin-section PCR was performed on each sample for HPV types and HIV. Comparisons were made between HPV types and groups of types identified in cancers.ResultsHPV DNA was identified in 92.0% of specimens from the U.S. contai… Show more

“…This finding, however, is higher than the report from a comparative study of HIV DNA rate in cervical tumor specimens of 31.5% in Botswana versus 3.9% in the United States. 15 The finding of high level of HIV coinfection correlates with the higher seroprevalence rate of 40% to 50% among females aged 30 to 50 years, 16 who comprised 52% of the study population. Several studies report variable HIVpositive seroprevalence among cervical cancer patients, varying from 1.6% in Hong Kong, 15% in Kenyas, and 21% in Durban, South Africa.…”

“…Acquisition of archived, paraffin-embedded blocks of formalin-fixed cervical cancer specimens from women residing in Indianapolis, Indiana (U.S.) and from Botswana was previously described [7]. Case selection was done by working backwards in time among archived paraffin blocks.…”

“…Thin-section polymerase chain reaction (PCR) of specimens from Kenyan women was performed exactly as described in a prior report of ICC cases from women living in Botswana and the U.S. [7]. Briefly, four to six paraffin-embedded tissue sections were scraped from slides prepared from each block using a sterile blade.…”
