Invariant Natural Killer T Cells and TGF-β Attenuate Anti-GBM Glomerulonephritis

Abstract: Invariant natural killer T (iNKT) cells represent a particular subset of T lymphocytes capable of producing several cytokines, which exert regulatory or effector functions, following stimulation of the T cell receptor. In this study, we investigated the influence of iNKT cells on the development of experimental anti-glomerular basement membrane glomerulonephritis (anti-GBM GN). After injection of anti-GBM serum, the number of kidney iNKT cells rapidly increased. iNKT cell-deficient mice (Jalpha18-/-) injected … Show more

“…8 Glomerulonephritis was induced in male C57BL/6 wild type (WT) or CalpTG mice as described previously. 23 Renal injury was evaluated on days 7 and 14. Specimens of kidney were cut for histochemistry and immunohistochemistry studies.…”

“…15 For the model of NTS-induced GN, decomplemented NTS was prepared as previously described. 37,38 Female wild-type and KO littermate mice of the C57BL/6 background (8-10 weeks old) received intravenous injections of 15 ml NTS per g body wt over two consecutive days (days 0 and 1) to induce crescentic GN. Control mice were injected with PBS.…”

“…Decomplemented NTS was prepared as previously described. 27 Female mice (aged 3 months) received intravenous injections of a total 30 ml of NTS/g body weight over 3 consecutive days (days 0, 1, and 2) to induce crescentic GN. Control mice were injected with PBS.…”
