Inulinase-hyperproducing strains of Kluyveromyces sp. isolated from aguamiel (Agave sap) and pulque

Abstract: Two strains of Kluyveromyces marxianus (A1 and A2) isolated from 'aguamiel' (agave sap) and one strain of K. lactis var. lactis (P7) isolated from 'pulque' (its fermented product), were studied to make a survey of inulinase production. The strains of K. marxianus A1 and A2 were the best producers of inulinase, giving up to 2.5 times more enzyme than the control hyperproducing strain K. marxianus CDBB-L-278, and showed lower catabolic repression than this. One strain isolated from pulque was identified as K. la… Show more

“…Many studies of K. marxianus have been carried out in liquid culture to investigate biomass or enzyme production [11][12][13][14][15][16][17][18]. However, although the potential applications are numerous, there have been few studies in solid-state culture (SSC), e.g., the production of inulinase [19], the production and optimisation of aroma compounds using factorial design and surface response methodology [20,21], and the production of inulinase and its preliminary characterization using factorial design [22,23].…”

“…Inulin and frutooligosaccharides (FOS) have proven bifidogenic effect (Kaur & Gupta, 2002;Ö zer, Akin, & Ö zer, 2005). ''Aguamiel'' is the sap obtained mainly from the species Agave atrovirens (Estrada-Godina et al, 2001) and contains a considerable amount of FOS (Cruz-Guerrero, Olvera, García-Garibay, & Gó mez-Ruiz, 2006), and can be considered as an interesting prebiotic and potential thermoprotector. The objective of this work was to demonstrate that B. bifidum viability following spray-drying and storage can be enhanced substantially by: (1) pre-selecting protective colloids that require high activation energy during their isothermal drying process (hinder oxygen diffusion through their drying matrices) as microencapsulating agents, and (2) by using aguamiel as thermoprotector prebiotic.…”

“…Pulque microflora have been described as containing mainly Lactobacillus spp., S. cerevisiae , Z. mobilis , and L. mesenteroides . Some other specific strains have also been isolated from this medium, such as Kluyveromyces marxianus , L. mesenteroides , and Leuconostoc kimchi . Valadez‐Blanco et al .…”
