Introduction of an efficient method for placenta decellularization with high potential to preserve ultrastructure and support cell attachment

Abstract: The placenta, as a large discarded tissue and rich in extracellular matrix (ECM), is an excellent candidate for biological scaffolds in reconstructive medicine.Considering the importance of ECM structure in cell fate, the aim of this study was to achieve human placenta decellularization protocol that preserve the structure of scaffolds. Thus, human placenta was decellularized by four protocols and decellularization efficacy was compared by hematoxylin and eosin (H&E), 4′,6-diamidino-2-phenylindole (DAPI) stain… Show more

“…In this study, the decellularization process was done using 1% SDS solution and 1% Triton X‐100. The efficacy of SDS and triton as detergents to decellularization tissue with high removal cells and minimum toxicity was reported in our published articles (Asgari et al, 2021; Azadbakht et al, 2023; Sajed et al, 2022). Decellularization method efficacy is usually investigated by histology staining and DNA content assessment in the samples, due to xenoantigenicity and proinflammatory reactions that can occur by DNA fragments (Gilpin & Yang, 2017).…”

“…4,5 Among the candidate tissues for decellularization, the human placenta is a frequently available source of tissue after birth. 6,7 They can be retrieved without undergoing an invasive operation, which gives them a significant advantage over other tissues and organs. The human placenta is a complex organ with a rich reservoir for various growth factors and cytokines such as epidermal growth factor (EGF), fibroblast growth factor (FGF), transforming growth factor-β (TGF-β), platelet-derived growth factor (PDGF), vascular endothelial growth factor (VEGF), etc.…”

“…8,9 ECM can be extracted and used from various cultured cells, and tissues, including human placenta tissue, which is normally discarded after birth. 10,11 The extracted ECM could be used as a biological component of the dressing to improve the healing. 12 This research aims to fabricate a hydrocolloid dressing functionalized with human placental ECM, which have optimum mechanical and biological properties.…”
