Introducing the Concept of Green Synthesis in the Undergraduate Laboratory: Two-Step Synthesis of 4-Bromoacetanilide from Aniline

Abstract: Incorporation of green chemistry techniques in undergraduate (UG) laboratories has a great demand. The challenge is to select and modify the known fundamental reactions in a greener way to implement into UG laboratories. A practical, two-step synthesis of 4-bromoacetanilide from aniline through a greener approach is described. Design of the synthetic route is based on the fundamental concepts of green chemistry. It can be adapted in most of the UG laboratories, including those with limited facilities. In the f… Show more

“…While in the previous method acetylation is carried out with corrosive acetic anhydride (LD 50 = 1.7 g/kg, rat, oral), Mukherjee and co-workers offer an attractive alternative method that is greener . They reported the acetylation of aniline using Zn dust and acetic acid (LD 50 = 3.3 g/kg, rat, oral).…”

“…PMI is a metric that compares the combined mass of all input materials in a process (reagents, solvents, separating agents, etc.) to the mass of isolated product obtained from the operative reaction. ,, It is selected as a comparative metric for this assignment as it is the preferred method used by the pharmaceutical industry to determine the material efficiency of reaction procedures . Students use the data provided in the assignment to complete the S N Ar PMI calculation and use the averaged experimental data from the laboratory to complete the B–H PMI calculation.…”

“…Green chemistry principles involve the use of safer alternatives to hazardous chemicals. Following the growing trend of green chemistry, many educators have striven to make introductory organic chemistry lab experiments more environmentally benign. Systems thinking influences green chemistry approaches and has informed much of the progress made in green chemistry . Systems thinking also encourages chemists to recognize the interdependence of chemistry with other disciplines and consider how they are related.…”
