Intranasal administration of oxytocin: Behavioral and clinical effects, a review

Abstract: The intranasal (IN-) administration of substances is attracting attention from scientists as well as pharmaceutical companies. The effects are surprisingly fast and specific. The present review explores our current knowledge about the routes of access to the cranial cavity. 'Direct-access-pathways' from the nasal cavity have been described but many additional experiments are needed to answer a variety of open questions regarding anatomy and physiology. Among the IN-applied substances oxytocin (OT) has an exten… Show more

“…Abnormalities in emotion recognition are central to numerous psychiatric conditions including ASD [96][97][98][99][100] and deficits in OT and insular cortex activity and connectivity are well documented correlates of symptom severity for many of these conditions 46,101,102 . Our results suggest that disruption of insula function may be key to pathophysiology in behaviors that depend upon social decisions regarding the emotions of others.…”

“…Similar to human (30) and recent nonhuman primate studies (31), the OT treatment was administered nasally, a minimally stressful method. One of the main technical issues of this method is whether the exogenous OT is able to cross the blood-brain barrier and exert direct effect on the brain.…”

“…administration has come to light as an effective and noninvasive means to circumvent the BBB for a variety of substances, including polypeptides, liposomes, virus vectors for gene therapy, and stem cells (Dhuria et al, 2010;Lochhead and Thorne, 2012a). These substances not only reach the brain tissue, but bioactive compounds such as insulin (Reger et al, 2008;Benedict et al, 2011;Craft et al, 2012), oxytocin (Veening and Olivier, 2013), and leptin (Schulz et al, 2012) do so in levels sufficient to cause significant behavioral changes in both patients and animal models of disease.…”
