Intragenic recombination in a flagellin gene: characterization of the H1-j gene of Salmonella typhi.

Abstract: Salmonella typhi, the etiologic agent of typhoid fever, typically has only a phase-1 flageliar antigen, d, but some isolates, found only in Indonesia, have antigenj instead, and may have a second flageliar antigen, z66. It appears that intragenic recombination involving a directly repeated 11 bp sequence in the HJ-d flagellin gene changed the flagellar antigen toj, by deleting 261 bp in its central, antigenically determinant, part. Sequencing of the hypervariable regions of genes HJ-d and Hi-j, and hybridizati… Show more

“…Alleles encoding flagellar antigens a, k, z 6 , z 10 , and z 41 were also found in loci different from that predicted by the Kauffmann-White scheme and different from where they were typically found. H:a is always listed in phase 1 in the KauffmannWhite scheme but was identified in fljB in serotype II 45:a:z 10 ( Table 3).…”

“…Alleles encoding several flagellar antigens were noted to contain deletions in what were otherwise genes encoding related flagellar antigens, including H:j, which has previously been reported to be encoded by an 86-amino-acid deletion in antigen H:d allele (10). Two alleles encoding H:z 40 were identical to alleles encoding H:l,v, except for a 19-or 7-amino-acid deletion in the variable region.…”

“…The variable nature of FliC and FljB flagellins together with the multiple lipopolyssacharide sidechains (O antigens), define more than 2,000 recognized Salmonella serovars (5). The natural diversity of Salmonella flagellins reflects mainly the polymorphism at the central portion of the protein, which creates sub-types such as illustrated by FliC flagellins expressed by S. Typhimurium (FliCi), S. Typhi (FliCd) and S. Muenchen (FliCd) (6).…”

“…This work has also shown that the population structure of the salmonellae is basically clonal, with each serovar being represented by one or a few predominant clones of widespread if not global distribution (Beltran et al, 1988 ;Reeves et al, 1989 ;Selander et al, 1990a, 6). Comparative sequencing of the flagellin genes in strains of several serovars has implicated both intragenic and interstrain recombination as mechanisms generating new serovars (Frankel et al, 1989;Smith & Selander, 1990;Smith et al, 1990).…”
