Abstract:Cytosolic free Ca2+ has been measured in living hyphae of Aspergillus awamori expressing a codon-optimised aequorin gene. Aequorin is a Ca2+-sensitive photoprotein, naturally produced by the jellyfish Aequorea victoria. Using this system we have found a range of treatments, which can be readily applied to fungi growing in 96 well plates, increase [Ca2+Ic. These include: high external CaC12 (0.01-100 mM); mechanical stimulation; and hypo-osmotic shock (but not hyper-osmotic shock). The Ca2+-
