Intracellular optical probing with gold nanostars

Abstract: The morphology of non-functionalized gold nanostars determines the surface protein structure and their interaction with the endolysosomal compartment in cells.

“…The combination of both types of information provides a broader picture of how the different cells process the nanostars. Differently from our previous work on fibroblast and epithelial carcinoma cells, where we have discussed the interaction of gold nanostars with the cells for long incubation times [ 23 ], here we will shed light on the influence of the composition of the biomolecular corona and the nanoparticle morphology, mainly spike length, for shorter incubation times that exclude late stages of endolysosomal maturation. As the data reported here demonstrate, nanostars taken up by 3T3 fibroblast cells, HCT-116 epithelial carcinoma cells, and J774 macrophages have a protein corona of different chemical and structural properties that are a consequence of the specific endolysosomal processing of each individual cell type.…”
