Intracellular Chloride Accumulation and Subcellular Elemental Distribution During Atrial Fibrillation

Abstract: Background-Ion channel remodeling occurs during atrial fibrillation (AF); however, the extent of alteration in the subcellular distribution of elements (Na, K, Cl, Ca, Mg, P) is unknown. Electron probe microanalysis was used to determine the total (freeϩbound) in vivo subcellular concentration of these elements during AF. Methods and Results-The left atrial appendage (LAA) was snap-frozen in situ after pacing (640 bpm) for 3 minutes (nϭ5 dogs), 30 minutes (nϭ3), or 48 hours (nϭ5). Dogs in sinus rhythm (nϭ3) se… Show more

“…In fact, after an initial increase, steady-state calcium levels begin to decrease as early as 30 minutes after initiation of AF and normalize within the first 48 hours. 27 Moreover, Ausma and coworkers 28 demonstrated decreased steady-state calcium levels after 16 weeks of AF in goats. Likewise, isolated (Figure 3).…”

“…An increase of intracellular chloride ions concentration was observed experimentally during atrial fibrillation in dogs [37], which was associated with ion channels remodeling during atrial fibrillation. An increased concentration of chloride in the extracellular environment (resulting from environmental pollution) could intensify this phenomenon.…”
