Intracellular calcium and pH conditions of cultured cells infected with Eimeria bovis or E. separata

Abstract: Loading of Eimeria bovis-infected Vero cells with membrane-permeant acetoxymethyl esters (AM-esters) of ion-sensitive dyes provided us with a noninvasive method for investigation of the permeability of the parasitophorous vacuole membrane (PVM) and simultaneous measurement of Ca2+ and H+ concentrations in different compartments of the infected cells. The distribution patterns of the cleaved membrane-impermeant dyes argue against the existence of nonselective pores in the PVM. There is also no indication of a p… Show more

“…Further, in their review, Saliba and Kirk (2000) discussed the possibility that the apparent difference in [Ca 2+ ] in the parasitophorous vacuole (PV) and host cell cytosol reflects other differences in composition between the two compartments. This is confirmed by findings of Behrendt et al (2000) who found the same [Ca 2+ ] in the host cell cytosol, the PV and the Fig. 5a, b The g t , Cl ) fluxes (J Cl ) and mannitol fluxes (J H ) of T. gondii-infected (5 and 15 h p.i.)…”

“…In fact, E. bovis sporozoite egress from cells may still occur later on observed it 8 days after infection], and can be provoked by mechanical irritation of the cell (e.g. during patch clamp experiments; Behrendt et al, unpubl.; see also Behrendt et al 2000). However, it happened generally only with parasites of the original slender type.…”

“…Thus, the quick development and expansion of the vacuole should be associated with an enhanced synthesis and secretion/exocytosis of such organellar products. Independent of these questions, however, the E. bovis-infected VERO cell with a large parasitophorous vacuole represents an interesting experimental tool, which has been recently used for permeability studies and analyses of Ca 2+ content and pH conditions in the various compartments of the infected cell (Behrendt et al 2000). Development of the parasite to mature schizont took place only in cells of bovine origin.…”

“…To the best of our knowledge, this is the first study to simultaneously measure force, [Ca 2ϩ ] i , and pH i in the portal vein. Previous studies in rat mesenteric vessels have shown the usefulness and validity of this approach (5), but few studies in any other tissue have been reported (3,8,20). The fluorescent spectra of the two indicators and the filters used were such that they did not interfere with each other.…”
