In vitro chromosome doubling can be induced by several antimitotic agents. The most commonly used are colchicine, oryzalin and trifluralin. The process of induced chromosome doubling in vitro consists of a typical succession of sub-processes, including an induction phase and a confirmation protocol to measure the rate of success. The induction step depends on a large number of variables: media, antimitotic agents, explant types, exposure times and concentrations. Flow cytometry is the pre-eminent method for evaluation of the induced polyploidization. However, alternative confirmation methods, such as chromosome counts and morphological observations, are also used. Since polyploidization has many consequences for plant growth and development, chromosome doubling has been intensively studied over the years and has found its way to several applications in plant breeding. This review gives an overview of the common methods of chromosome doubling in vitro, the history of the technique, and progress made over the years. The applications of chromosome doubling in a broader context are also discussed.

The aim of the research was to establish an efficient procedure for in vitro micropropagation in order to obtain numbers of identical plants for in vitro polyploidization of Humulus lupulus (2n = 20), using antimicrotubule agent oryzalin. For this purpose, the polyploidization was carried out for H. lupulus Osvald’s clones 31, 74, 114, and for ‘Sladek’ cultivar. The two experimental methods – the cultivation of nodal segments on medium with different concentrations of oryzalin (1, 5, and 10 µM) for 2 weeks and the irrigation of nodal segments with oryzalin (10 and 20 µM) for 24 and 48 h were chosen for inducing for polyploid plantlets of H. lupulus. This procedure provided tetraploids, which were identified by flow cytometry using internal standardization method and confirmed using chromosome counting of methaphasic cells from the root tips and morphological observations. The influence of chromosome doubling was also verified with stomata characterization. The polyploid plants were propagated for next evaluation, rooting and transfer to nonsterile conditions and into field. After chromosome doubling, using some different concentration of oryzalin, some plantlets became tetraploids, no mixoploids were detected. The highest efficiency of polyploidization was achieved for clone 72 after 2-week treatment of oryzalin supplemented medium. On the other hand, for method based on the irrigation of nodal segments with oryzalin, the most efficient conditions were treatment with 10 µM and 20 µM oryzalin for 24 and 48 h, respectively.

A protocol for in vitro induction of tetraploids of the grape rootstock 101-14 Millardet et de Grasset (Vitis riparia 9 V. rupestris) (101-14 Mgt) (2n = 2x = 38) 9 Muscadinia rotundifolia cv. Trayshed (2n = 2x = 40) hybrids was established. Different explant materials were exposed to the antimitotic chemical agents colchicine and oryzalin, at three concentrations, for 24, 48 and 72 h. Treated in vitro shoot tips, anthers and pre-embryogenic cultures were regenerated, and their ploidy levels were confirmed by chromosome counts and flow cytometry. It is generally considered that anatomical characteristics of leaves, including leaf stomatal size and number of chloroplasts in guard cells, correlate with ploidy levels of grape plants. Chromosome counts and flow cytometry are labor intensive and difficult, while the number of chloroplasts in stomatal guard cells and stomatal size are easily measured and can be stable and reliable markers. Analysis of stomata revealed clear differences between regenerated diploid and tetraploid plants, and these measurements were used as a preliminary screen, followed by chromosome counts and flow cytometry for confirmation. This study presents the results obtained by various combinations of explants, antimitotic agents, exposure time, concentration and ploidy analysis to evaluate in vitro chromosome doubling. These results provide an easy and effective method for production of tetraploid, potentially fertile Vitis 9 Muscadinia hybrids for utilization in rootstock breeding programs.
