Intra‐ and inter‐specific crosses of Solanum materials after mitotic polyploidization in vitro

Abstract: Mitotic polyploidization in vitro was used in selected wild Solanum species and Solanum tuberosum dihaploids. The efficiency of polyploidization by colchicine was compared with that of oryzalin. Oryzalin was more effective than colchicine (P ¼ 0.1). The rate of non-affected to mixoploid to tetraploid regenerants was 22 : 2.5 : 1 (colchicine) and 14 : 2 : 1 (oryzalin). Optimal concentrations and durations were 3.5 mM/24 h for colchicine and 25 or 30 lM for 24 or 48 h for oryzalin (variations in concentration an… Show more

“…It contributes to the effect and toxicity of the treatment. In many studies, antimitotic agents are dissolved in DMSO (Allum et al 2007;Awoleye et al 1994;Greplova et al 2009;Hamill et al 1992;Kermani et al 2003;Khosravi et al 2008;Stanys et al 2006;Subrahmanyam and Kasha 1975;Takamura et al 2002;Väinölä 2000;Yang et al 2006;Zhang et al 2007Zhang et al , 2010a. DMSO increases cell permeability and allows increased absorption of chemicals (Hamill et al 1992).…”

“…Nodal segments (third to fifth from the apex of donor plants) with one leaf were used as explants. A stock solution of oryzalin (Sigma-Aldrich, St. Louis, MO, USA) was prepared according to Greplová et al [27] and the respective concentrations of oryzalin for the experiments were prepared from this stock solution.…”

“…More recently, herbicides such as oryzalin, which also interfere with plant cell tubulins, have been used as a mitotic blocking agent for polyploidization because of its reduced toxicity for humans (Dhooghe et al 2011). The artificial induction of polyploidy with oryzalin has been employed successfully in fruit species such as Malus (Bartish et al 1999), Musa (Ganga and Chezhiyan 2002;Van Duren et al 1996), Smallanthus sonchifolius (Viehmannová et al 2009) and Solanum (Greplova et al 2009). …”
