Intestinal HIF-2α Regulates GLP-1 Secretion via Lipid Sensing in L-Cells

Abstract: Activation of intestinal hypoxia-inducible factor signaling improves glucose homeostasis in a glucagon-like peptide-1dependent manner. We show that activation of intestinal hypoxia-inducible factor 2a signaling increases GLP-1 levels and attenuates diet-induced metabolic perturbations such as visceral adiposity, glucose intolerance, and hepatic steatosis.

“…At week 4, we performed intraperitoneal glucose tolerance test (GTT) and insulin tolerance test (ITT) in mice respectively, according to the methods described by Le, J 35 and Mooli. 36 Mice were placed in clean cages and fasted with free access to water. For the GTT, mice were fasted for 12 h and baseline blood glucose was measured using a glucometer.…”

“…Cells or human and mouse frozen tissues (~10mg) were homogenized using radioimmunoprecipitation assay lysis buffer (0.5% NP-40, 0.1% sodium deoxycholate, 150 mmol/L NaCl, 50 mmol/L Tris-Cl, pH 7.5) containing 1 mmol/L phenylmethylsulphonyl fluoride, protease inhibitor cocktail (Sigma-Aldrich) and 2 mmol/L sodium orthovanadate 24 . The homogenate was centrifuged at 13,000 × g for 10 min at 4°C to collect the supernatant.…”

“…The authors showed that activation of intestinal HIF2α during obesity increased ceramide synthesis fueling hepatic steatosis. However, Mooli et al (81) reported that intestinal HIF2α regulated lipid sensing and increased GLP-1 secretion, thereby protecting against diet-induced obesity and hepatic steatosis. These contradictory findings suggest a complex role for intestinal HIF2α influencing hepatic steatosis, which may further be attributed to activation of HIF2α in a specific intestinal cell type, enterocytes, compared with enteroendocrine L cells.…”
