Intestinal hephaestin potentiates iron absorption in weanling, adult, and pregnant mice under physiological conditions

Abstract: Key Points Intestinal Heph may be required to potentiate iron absorption during rapid growth and pregnancy, or when hypoxia stimulates erythropoiesis. Here, the physiological conditions in which Heph is required are defined and other, complementary, intestinal ferroxidases are identified.

“…Figure 2 also shows that the cell viability of iron-based implant was lower than the cell viability of stainless steel implant using hemocytometer ( Figure 2(a) ) and MTT assay ( Figure 2(b) ). Iron is a chemical element that is active in the fundamental physiologic processes [ 12 ], but a higher concentration of iron could induce cellular apoptosis [ 19 ]. Therefore, the cell motility rate in iron-based implant was higher.…”

“…Iron has attractive mechanical properties, approaching those of stainless steel. Due to the formation of a complex iron phosphate layer, iron has a low degradation rate [ 11 ] and is required in little quantity [ 12 ]. A lot of iron-based implants research has been conducted, but just a few of them discussed the potential of the iron-based implant in a clinical study chronically.…”

“…Mice were anesthetized by CO 2 exposure and killed by cervical dislocation. Hemoglobin (Hb) and hematocrit (Hct) levels in whole blood were determined by standard methods [ 18 ]. Plasma non-heme iron levels were measured using a colorimetric method [ 19 ].…”

“…Intestinal iron absorption and body iron distribution in experimental mice were determined after oral administration of 59 Fe, as previously described [ 18 ]. Briefly, mice were provided their final dose of saline, or FA-GDLVs/siRNA, and then three hours later, food was removed from cages (but mice were allowed free access to water during this time).…”
