Interspecific Recombination between Two Ruminant Alphaherpesviruses, Bovine Herpesviruses 1 and 5

Abstract: Homologous recombination between different species of alphaherpesviruses has been described between herpes simplex viruses 1 and 2 but has not yet been observed between other alphaherpesviruses. In the present study we chose to assess to what extent in vitro recombination can occur between members of a well-defined group of closely related viruses such as ruminant alphaherpesviruses. At 24 h after infection of epithelial bovine kidney cells with a double-deleted mutant of bovine herpesvirus 1 (BoHV-1) (contain… Show more

“…On the other hand, interspecies recombinants have been regarded to survive rarely in nature (5). Although there are some reports on interspecies recombinants of closely related herpesvirus species, such as HSV-1 and HSV-2 (6, 13, 36) and BHV-1 and BHV-5 (20), in laboratory, no natural recombinant has been detected. Exceptional examples have been reported on HHV-8, some strains of which seemed to be descendants of an interspecies recombinant (25).…”

“…Indeed, successful homologous recombination requires two closely related parental viruses. Two recombinants between BoHV-1 and BoHV-5 have been isolated, but no recombinant has been detected between BoHV-1 and the less closely related CpHV-1 and CvHV-2 [78]. Nevertheless, even in the absence of mature recombinant viruses, these results do not exclude a recombination process occurring in the cell, because mixed concatemers between BoHV-1 and CvHV-2, and between CvHV-1 and CvHV-2, have been identified in co-infected cells 3 .…”

“…Restriction endonuclease analysis (REA) performed on viral isolates allows the identification and distinguishing of the related viruses [12, 14, 15, 23, 38-41, 94, 98, 100, 103, 137, 145] and could be used to detect interspecific recombinants [78]. However, this procedure is not convenient for general use in a diagnostic laboratory.…”

“…It is most likely coupled with viral DNA replication and may require both viral and cellular factors (4,11,43). Homologous recombination can occur only between closely related genomes (16,27). Nonetheless, the creation of new genomes requires some sequence heterogeneity in the genomes involved in recombination.…”
