Interplay of SOX and POU Factors in Regulation of the Nestin Gene in Neural Primordial Cells

Abstract: Intermediate-filament Nestin and group B1 SOX transcription factors (SOX1/2/3) are often employed as markers for neural primordium, suggesting their regulatory link. We have identified adjacent and essential SOX and POU factor binding sites in the Nestin neural enhancer. The 30-bp sequence of the enhancer including these sites (Nes30) showed a nervous system-specific and SOX-POU-dependent enhancer activity in multimeric forms in transfection assays and was utilized in assessing the specificity of the synergism… Show more

“…Digoxigenin-labeled antisense riboprobes for Sip1 and Sox2 were prepared using full-length Sip1 cDNA sequence ( Van de Putte et al, 2003) and the 533-bp SacII-AccI fragment of Sox2 (Tanaka et al, 2004), respectively. Whole mount in situ hybridization was carried out as described by Wilkinson (1992) with the following modifications: incubation with glycine after Proteinase K treatment was omitted; anti-digoxigenin antibody was reacted in 1.5% Blocking Reagent (Roche) instead of 10% sheep serum.…”

“…Mouse monoclonal antibodies included: anti-GFP (1:400, MP Biomedicals), anti-Oct4 (1:200, Santa Cruz Biotechnology), anti-nestin (1:200, Upstate), anti-Tuj1 (1:500, Sigma), anti-MAP2 (1:500, Sigma), anti-AP2γ (1:500, Abcam), anti-cytokeratin18 (1:200, Abcam) and anti-cytokeratin14 (1:50, Abcam). Rabbit polyclonal antibodies were anti-GFP (1:400, Santa Cruz Biotechnology), an anti-Sox1/(2)/3 (1:100) that has a preference for Sox1 and Sox3 over Sox2 [78,79]. Fluorescein isothiocyanate-and Cy3-conjugated secondary antibodies were used (1:200 and 1:500, Jackson ImmunoResearch).…”

“…136 Nestin expression is associated with cell migration and metastasis in prostate cancer, 137 and tumor progression and deceased survival in melanoma. 41,[138][139][140][141][142] Nestin and SOX2, 143 an embryologic stem cell transcription factor that binds an enhancer region on the nestin gene, 144 are preferentially coexpressed in metastatic melanomas when compared with nevi or primary melanomas. 145 Moreover, SOX2-positive melanoma cells tend to be more spindleshaped cells and to have a more peripheral nestin pattern, which may represent a motile, more mesenchymal phenotype.…”
