Interplay of negative and positive signals controls endoderm-specific expression of the ascidian Cititf1 gene promoter

Abstract: Cititf1 is an early and specific marker of endoderm development in Ciona intestinalis [ Development 126, 5149]. Here, we examine Cititf1 transcriptional regulation focusing in particular on its endodermal restricted expression. Through the analysis of Ciona embryos, electroporated with different portions of Cititf1 5'-flanking region fused to lacZ, we characterized a minimal 300-bp cis-regulatory sequence able to closely reproduce the spatial and temporal expression pattern of the endogenous gene. This enhance… Show more

“…EMSA assay has been performed as previously reported (Fanelli et al, 2003), using 5.0 l of in vitro synthesized Ci-Ets1/2 DBD protein, and labeled and competitor DNAs prepared by annealing the wildtype (Wt2) or mutant (Mut2) oligonucleotides (see Table S3 in the supplementary material).…”

“…Such approaches are now being applied to the early development of the heart (B. Davidson, W. Shi, and M. Levine, in prep. ), endoderm (Satou et al 2001;Fanelli et al 2003;Imai 2003;Oda-Ishii et al 2005), tail muscles (Yagi et al 2005), and stomodaeum (Christiaen et al 2005). In combination with the comprehensive gene-expression studies and functional assays discussed previously, these enhancer studies will set the stage for deciphering the basic gene networks in the Ciona embryo.…”

“…cis-Regulatory regions responsible for the endoderm specific expression of Cititf1 have been isolated and characterized (Ristoratore et al, 1999;Fanelli et al, 2003). The 315-bp genomic fragment spanning from -355 to -41 relative to the transcription start site is sufficient to activate a heterologous promoter in a pattern essentially identical to that of the endogenous Cititf1 gene (Fanelli et al, 2003).…”

The 315-bp genomic fragment spanning from -355 to -41 relative to the transcription start site is sufficient to activate a heterologous promoter in a pattern essentially identical to that of the endogenous Cititf1 gene (Fanelli et al, 2003). This enhancer contains at least three distinct regulatory regions; two of which are responsible for activation of transcription in the endoderm and in the mesenchyme, respectively, while the third is a negative control region that represses transcription in mesenchyme.…”
