Interplay between integrins and FLK-1 in shear stress-induced signaling

Abstract: We studied interactions between integrins and Flk-1 in transducing the mechanical shear stress due to flow. This application of a step shear stress caused Flk-1 ⅐ Casitas B-lineage lymphoma (Cbl) activation (Flk-1 ⅐ Cbl association, tyrosine phosphorylation of the Cbl-bound Flk-1, and tyrosine phosphorylation of Cbl) in bovine aortic endothelial cells (BAECs). The activation of integrins by plating BAECs on vitronectin or fibronectin also induced this Flk-1 ⅐ Cbl activation. The shear-induced Flk-1 ⅐ Cbl activ… Show more

“…Caveolae have been shown to be an essential component of the pathway for EC signaling associated with abrupt reduction of shear stress [29]. It was reported integrins are essential for the mechanical activation of Flk-1 by SS but not for the chemical activation of Flk-1 by VEGF [6]. The signaling events induced by SS and VEGF converge at the membrane receptor Flk-1 and that these stimuli share the Flk-1/Cbl/Akt pathway in activating IKK activation [7].…”

“…Hence, a Flk‐1‐mediated, ligand‐independent mechanotransduction may contribute more significantly to this form of angiogenesis than VEGF per se (Wang et al . 2002, Baum et al . 2004), offering a parallel with the situation in low‐flow ischaemia (Milkiewicz et al .…”

“…The effects of shear stress on endothelial cell integrins include their clustering [52], association with the adaptor protein Shc [53], and binding with WOW-1, which is an antibody that specifically recognizes the activated intergrin v 3 [54]. The shear-induced activation involves dynamic and specific interactions between integrins and their cognate extracellular matrix ligands [55].…”
