Internal/Structures Containing Transcriptase-Related Proteins in Top Component Particles of Mammalian Orthoreovirus

Abstract: The structure of mammalian orthoreovirus top component particles, which are profoundly deficient in the content of double-stranded RNA genome, was determined at 30 A resolution by transmission cryoelectron microscopy and three-dimensional image reconstruction. Previously undetected, ordered densities, appearing primarily as pentameric flowers in the reconstruction, were seen to extend 65 A inwardly from the inner capsid at the icosahedral fivefold axes. Identically positioned but lower density elements were ob… Show more

“…4c,d). The broad interface between λ3 and λ1 suggests that λ3 is firmly anchored to the shell, consistent with the 'fixed-polymerase-movingtemplate' model of reovirus transcription 15,18,44,45 . Despite the inherent high noise level in the central parts of the cryo-EM map, 33 of the 48 α-helices in the λ3 X-ray crystal model were clearly identified and fitted by both SITUS and EMfit (Table 1).…”

“…However, in none of these viruses have the positions and orientations of the RdRps been precisely determined. For all these viruses, a generally accepted, though largely unproven, model is that each of the 10-12 genome segments is dedicated to its own transcriptase complex within the particle 15,18,31,[35][36][37][38][39] .…”

“…Nascent mRNA is capped before being released from the intact capsid. The mechanism by which this characteristic endogenous transcription takes place inside intact dsRNA viruses has been studied extensively (3)(4)(5)(6)(7), and it is generally believed that the RNA polymerase complex is structurally anchored to the core during transcription (7)(8)(9). The dsRNA template must be flexible enough to move freely within the densely packed core so that it can slide through the RNAdependent RNA polymerase (RDRP) and the capping enzyme complex to ensure efficient transcription (10).…”

“…We present results regarding two virus structures, those of Sindbis and reo viruses [10]. The experimental data for the Sindbis virus consist of 7,917 views of 221 × 221 pixels each; the highest resolution obtained using existing algorithms is 11.2 Å.…”
