Interleukin-8 in sepsis: relation to shock and inflammatory mediators

Abstract: Because of its neutrophil-activating properties, interleukin-8 (IL-8) may play an important role in the pathophysiology of sepsis. We measured circulating IL-8 levels in 47 patients with clinical sepsis. Levels on admission were elevated in 42 of the 47 patients (89%o) and were comparable in patients with gram-positive or gram-negative infections. Patients with shock had significantly higher IL-8 levels than normotensive patients (P = 0.0014, Wilcoxon-Mann-Whitney test), whereas no differences in IL-8 levels w… Show more

“…Serum was obtained by centrifugation at 4 C for 20 min at 1600 g, and stored at biotinylated anti-IL-10 MoAb JES3-12G8 (Pharmingen) as detecting antibody, and recombinant human IL-10 (Medgenix, Fleurus, Belgium) as standard, exactly as described previously [9]. IL-6 and IL-8 were measured by ELISA [15,16].…”

“…Minimum detection level for sGranzyme A was 3 pg/ml and for sGranzyme B 2 pg/ml. IL-8 [18], IL-10 [19], IL-12p40, IL-12p70, IFN g [20] and CRP [21] were also measured by ELISA, accordingly the publication procedure.…”

“…CLB-FcR/gran-1 (5D2, IgG2a [16]) was produced in our own institute. The MoAbs against tumour necrosis factor-alpha (TNF-a) (aTNF/5, aTNF/7 [17]) and IL-8 (aIL-8/1 [18]) have been described before. F(ab) fragments were made by digestion with 4% (w/w) papain for 1·5 h at 37ЊC in PBS containing 10 mM cysteine and 5 mM EDTA.…”
